Supported by their involvement in many neurodegenerative disorders, muscarinic acetylcholine receptors ( mAChRs) are an interesting target for PET imaging. Nevertheless, no radiotracer is established in clinical routine. Within this work we aim to develop novel PET tracers based on the structure of arecoline. Fifteen novel arecoline derivatives were synthesized, characterized and tested for their affinity to the mAChRs M1-M5 and the conceivable off-target acetylcholinesterase. Five arecoline derivatives and arecoline were labeled with carbon-11 in good yields. Arecaidine diphenylmethyl ester (3b), arecaidine bis(4-fluorophenyl)methyl ester (3c) and arecaidine (4-bromophenyl)(4-fluorophenyl)methyl ester (3e) showed a tremendous gain in mAChR affinity compared to arecoline and a pronounced subtype selectivity for M1. Metabolic stability and serum protein binding of [C-11] 3b and [C-11] 3c were in line with properties of established brain tracers. Nonspecific binding of [C-11]3c was prevalent in kinetic and endpoint experiment on living cells as well as in autoradiography on native mouse brain sections, which motivates us to decrease the lipophilicity of this substance class prior to in vivo experiments. Nucleophilic substitutions of benzylic alcohols with sulfamides were achieved using an FeCl3 Lewis acid catalyst in MeNO2. It was necessary to adjust the reaction conditions to obtain efficient yields depending on the stability of the carbocation intermediates. The reaction could easily be performed, and it was revealed that a variety of diarylmethanols and benzylic alcohols were applicable to the reaction, irrespective of the type and position of the substituents. The sulfamide moieties were easily deprotected and converted into amine groups.

A series of oxy-ether tris-amino heteroditopic macrobicycles (L1-L4) with various cavity dimensions have been synthesized and explored for their Cu(II) catalyzed selective single step aerial oxidative cross-coupling of primary alcohol based anilines with several aromatic amines toward the formation of primary alcohol appended cross azobenzenes (POCABs). The beauty of this transformation is that the easily oxidizable benzyl/primary alcohol group remains unhampered during the course of this oxidation due to the protective oxy-ether pocket of this series of macrobicyclic vessels. Various dimensionalities of the molecular vessels have shown specific size complementary selection for substrates toward efficient syntheses of regioselective POCAB products. To establish the requirement of the three-dimensional cavity based additives, a particular catalytic reaction has been examined in the presence of macrobicycles (L2 and L3) versus macrocycles (MC1 and MC2) and tripodal acyclic (AC1 and AC2) analogous components, respectively. Subsequently, L1-L4 have been extensively utilized toward the syntheses of as many as 44 POCABs and are characterized by different spectroscopic techniques and single crystal X-ray diffraction studies.

In one-pot strategy, diazotization of methyl anthranilate 5 followed by addition of amino acid ester hydrochloride, we have prepared methyl-2-(4-oxobenzotriazin-3(4H)-yl)-alkanoates 6a-c. Starting with hydrazides 7a,b, N-alkyl-2-(4-oxobenzotriazin-3(4H)-yl)-alkanamides 9-10(a-h) and methyl-2-(2-(4-oxobenzotriazin-3(4H)-yl)alkanamido)alkanoates 11- 12(a-e) were prepared via azide coupling. Hydrazones 13-15 were prepared via condensation of hydrazides 7a,b with 4-methoxybenzaldehyde, 4-dimethylaminobenzaldehyde, and/or arabinose. Molecular docking was done for synthesized compounds using MOE 2008-10 software. The compounds 9a, 12a, 12c, 13a, 13b, and 14b have the most pronounced strong binding affinities toward the target E. coli Fab-H receptor, whereas compounds 3, 11e, 12e, and 13a have the most pronounced strong binding affinities toward the target vitamin D receptor. The in vitro antibacterial activities of the highest binding affinity docked compounds were tested against E. coli, Staphylococcus aureus, and Salmonella spp. Majority of the tested compounds showed effective positive results against E. coli, while they were almost inactive against Staphylococcus aureus and Salmonella spp. The in vitro cytotoxic activities of the highest binding affinity-docked compounds were tested against the human liver carcinoma cell line (HepG2). Some compounds showed potent cytotoxic activity with low IC50 values, especially for 3 (6.525 mu M) and 13a (10.97 mu M) than that for standard drug doxorubicin (2.06 mu M).

The bifunctional ligand p-SCN-Bn-HOPO, which has four 1,2-hydroxypyridinone groups on a spermine backbone with an isothiocyanate linker, has been shown to be an efficient and stable chelator for Zr(iv) and, more importantly, the radioisotope Zr-89 for use in radiolabeling antibodies for positron emission tomography (PET) imaging. Previous studies of Zr-89-HOPO-trastuzumab in mice showed low background, good tumor to organ contrast, and very low bone uptake which show p-SCN-Bn-HOPO to be an important next-generation bifunctional chelator for radioimmunoPET imaging with Zr-89. However, the reported synthesis of p-SCN-Bn-HOPO involves nine steps and multiple HPLC purifications with an overall yield of about 1.4%. Herein we report an improved and efficient synthesis of p-SCN-Bn-HOPO in four steps with 14.3% overall yield which will improve its availability for further biological studies and wider application in PET imaging. The new synthetic route also allows variation in linker length and chemistries which may be helpful in modifying in vivo clearance behaviors of future agents.

Herein we report the iron-catalyzed beta-C(sp(3))-methylation of primary alcohols using methanol as a Cl building block. This borrowing hydrogen approach employs a well-defined bench-stable (cyclopentadienone)iron(0) carbonyl complex as precatalyst (5 mol %) and enables a diverse selection of substituted 2-arylethanols to undergo beta-C(sp(3))-methylation in good isolated yields (24 examples, 65% average yield).

Perylenediimides (PDIs) have emerged as potential materials for optoelectronic applications. In the current work, four PDI derivatives, substituted at imide nitrogen with 2,6-diisopropyl phenyl, 2-nitrophenyl, diphenylmethylene, and pentafluorophenyl groups, have been synthesized from perylene 3,4,9,10-tetracarboxylic dianhydride using a facile imidization synthesis process. PDI derivatives have been spectroscopically characterized for their structure and optical properties. Temperature-variable absorption and emission spectroscopy study confirmed the H-aggregation property. H-aggregation along with strong emission suggests the slipped pi-pi stacking of PDI molecules. Electrochemical analysis was performed for their redox behavior and calculation of lowest unoccupied molecular orbital and highest occupied molecular orbital energy levels. Scanning electron microscopy showed the formation of ordered structures. The PDI derivatives showed excellent electron conductivity without doping and 5-10x higher electron mobility than that of state-of-the-art fullerene acceptor phenyl-C-61 -butyric acid methyl ester (PC61BM). Finally, the charge generation and charge transfer phenomenon was studied by transient absorption spectroscopy (TAS). TAS showed ultrafast charge transfer from the poly(3-hexyl)thiophene (P3HT) donor polymer to PDI and formation of long-lived charge-separated states similar to fullerene derivative PC61 BM/P3HT blends. Such PDI derivatives with excellent solubility and photophysical and electronic properties are potential n-type materials to be used in organic electronic devices.

Based on the characteristic of benzo[d][1,2,3]thiadiazole to induce the systemic acquired resistance and improve the immunity of plants, benzo[d][1,2,3]thiadiazole was introduced into 1,2,3-benzotriazin-4-one, thirty-one novel 1,2,3-benzotriazin-4-one derivatives containing benzo[d][1,2,3]thiadiazole were designed and synthesized. Nematicidal activity showed that most of the synthesized compounds exhibited great inhibitory activity in vivo against Meloidogyne incognita at 20 mg/L. Among 31 tested compounds, A2 and A3 showed an excellent nematicidal activity with the inhibition rate of 50.4% and 53.1% at the concentration of 1.0 mg/L, respectively. The influence of substituent type and position was investigated. The relationship between structure and activity was also preliminary analyzed.

Malaria, despite many efforts, remains among the most problematic infectious diseases worldwide, mainly due to the development of drug resistance by Plasmodium falciparum. The antibiotic fosmidomycin (FSM) is also known for its antimalarial activity by targeting the non-mevalonate isoprenoid synthesis pathway, which is essential for the malaria parasites but is absent in mammalians. In this study, we synthesized and evaluated against the chloroquine-resistant P. falciparum FcB1/Colombia strain, a series of FSM analogs, derivatives, and conjugates with other antimalarial agents, such as artemisinin (ART) and aminochloroquinoline (ACQ). The biological evaluation revealed four new compounds with higher antimalarial activity than FSM: two FSM-ACQ derivatives and two FSM-ART conjugates, with 3.5-5.4 and 41.5-23.1 times more potent activities than FSM, respectively.
