Background: Increased cigarette costs have inadvertently strengthened the appeal of discounted brands to price-sensitive smokers. Although smokers perceive discounted brands as having poorer quality, little is known about their delivery of toxic tobacco smoke constituents compared with premium-branded tobacco products. Methods: We investigated the differences between discount and premium brand smokers using the National Health and Nutrition Examination Survey 2011-2012 Special Smoker Sample. Our analyses focused on demog. differences and 27 biomarkers of harmful and potentially harmful constituents (HPHC) listed by the FDA, including volatile organic compounds, 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol and its glucuronide [4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol glucuronide; reported as total NNAL (tNNAL)], metals, and polycyclic aromatic hydrocarbons (PAHs). Data were analyzed using linear regression models adjusting for potential confounders. Results: A total of 976 non-tobacco users and 578 recent cigarette smokers were eligible for anal., of which 141 (26.0% weighted) smoked discount brand cigarettes and 437 (74.0% weighted) smoked premium. Discount brand smokers were older, predominantly non-Hispanic white, and had higher serum cotinine. Discount brand smokers had significantly higher levels of 13 smoking-related biomarkers, including tNNAL, uranium, styrene, xylene, and biomarkers of exposure to PAHs (naphthalene, fluorene, and phenanthrene), compared with premium brand smokers. Conclusions: These findings suggest that discount cigarette use is associated with higher exposure to several carcinogenic and toxic HPHCs. In microbiol. methods, the data obtained from the analyzes performed to determine the potency of antibiotics are the inhibition zone diameters or turbidimetric turbidity data. In this work, aqueous phase recirculation was conducted in the ethanol-water solvent to evaluate the feasibility of ethanol addition in inhabiting the re-polymerization of nitrogenous compounds The present study provides a comprehensive comparison of yield and quality between bio-oils derived from aqueous phase recirculation in the water solvent and ethanol-water solvent. The results showed that the addition of ethanol in aqueous phase recirculation displayed higher promotion in bio-oil yield (7%) compared with water solvent (3.55%). The energy recovery efficiency of bio-oil from ethanol-water solvent reached 87.43% after recirculation for three times, while that from the water solvent was 74.61%. The elemental anal. and gas chromatog.-mass spectrometry anal. indicated that ethanol addition could inhibit the re-polymerization of nitrogenous compounds and thus decrease the nitrogen content in bio-oil. This resulted in lower NO and HCN emission peaks during the combustion process. Besides, the addition of ethanol increased the content of light mol. weight fractions and decreased the coke formation, which contributed to complete combustion and resulted in lower CO emission peaks. Overall, this work provides a feasible way for further improving the bio-oil yield and fuel quality through ethanol addition in solvent during aqueous phase recirculation. Thus, Kin at 3.11 渭M allowed high regeneration of vitroplants, optimal elongation, total rooting of explants, maximum bud multiplication, and absence of hyperhydric explants. In fact, the integration of auxins (indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), and 1-naphthaleneacetic acid (NAA)) into the culture medium and their combinations with 3.11 渭M Kinetin contributed to the optimization of the root part. Thus, it was improved in particular in the case of 3.11 渭M Kin and 6.27 渭M IBA. Three polyamines (Putrescine, Spermidine and Spermine) at different concentrations (1.134, 3.402, 5.67, 7.938 and 11.34 渭M/L) combined at 3.11 渭M Kin and 6.27 渭M IBA were tested. In fact, 1.304 渭M putrescine was considered to be the most suitable for in vitro culture of explants, since it allowed optimal propagation of buds and roots, also a high rate of regeneration and rhizogenesis. The acclimatization was carried out successfully using vitroplants showing good foliar and root development. Abstract: Phytohormones, such as auxins and cytokinins, take part in the integration of growth control and stress response, but their role in algal adaptation to heavy metal remains to be elucidated. The current research indicated that lead (Pb), one of the most toxic metals in nature, causes severe depletion of endogenous cytokinins, auxins, and gibberellin and an increase in abscisic acid content in the green alga Acutodesmus obliquus. Exogenous auxins and cytokinins alleviate Pb toxicity through the regulation of the endogenous phytohormones鈥?levels. Exogenously applied auxins provoked the coordinated activation metal tolerance mechanisms leading to the increase in phytochelatin synthase activity and accumulation of phytochelatins and their precursors, which are essential for Pb sequestration. On the other hand, phytochelatin synthesis decreased in algal cells treated with cytokinins. Significant changes in the levels of low mol. weight metabolites, mainly involved in metal chelation and glutathione synthesis pathway under the influence of phytohormones in algal cells growing in the presence of Pb stress, were observed This is the first report showing that auxins and cytokinins are important regulatory factors in algal adaptation strategies to heavy metal stress based on thiol-mediated compounds and the maintenance of phytohormone homeostasis. We sought to design ubiquitin-proteasome system inhibitors active against solid cancers by targeting ubiquitin receptor RPN13 within the proteasome鈥瞫 19S regulatory particle. The prototypic bis-benzylidine piperidone-based inhibitor RA190 is a Michael acceptor that adducts Cysteine 88 of RPN13. In probing the pharmacophore, we showed the benefit of the central nitrogen-bearing piperidone ring moiety compared to a cyclohexanone, the importance of the span of the aromatic wings from the central enone-piperidone ring, the contribution of both wings, and that substituents with stronger electron withdrawing groups were more cytotoxic. Potency was further enhanced by coupling of a second warhead to the central nitrogen-bearing piperidone as RA375 exhibited ten-fold greater activity against cancer lines than RA190, reflecting its nitro ring substituents and the addition of a chloroacetamide warhead. Treatment with RA375 caused a rapid and profound accumulation of high mol. weight polyubiquitinated proteins and reduced intracellular glutathione levels, which produce endoplasmic reticulum and oxidative stress, and trigger apoptosis. RA375 was highly active against cell lines of multiple myeloma and diverse solid cancers, and demonstrated a wide therapeutic window against normal cells. For cervical and head and neck cancer cell lines, those associated with human papillomavirus were significantly more sensitive to RA375. While ARID1A-deficiency also enhanced sensitivity 4-fold, RA375 was active against all ovarian cancer cell lines tested. In this work, a chip, composed of high-d. Si nanopillars and Ag nanoislands, is prepared for sensitive detection of sulfonamides with surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS). In this chip, the high-d. Si nanopillars can absorb laser energy efficiently and promote electron transfer as antennas; the Ag nanoislands can enhance the thermal effect; the formation of the Schottky barrier and the conductive nanolayer between Si/Ag can extend the lifetime of electrons and promote electron tunneling effect. Therefore, the heterostructure presents high detection sensitivity (the limit of detection of sulfamethazine is as low as 3 amol); in addition, the satisfactory coefficients of determination (> 0.99) show that the chip is applicable for rapid quantification of sulfonamides. Further, the sulfacetamides spiked in milk and lake water can be sensitively identified and rapidly quantified.
