Piper nigrum (black pepper), Piper longum (Indian tipali) and Piper chaba (Bangla tipali) are widely used medicinal herbs in traditional Indian systems of medicine. P. longum as well as P. chaba are considered to be inferior substitutes of P. nigrum. A rapid anal. method for metabolic profiling was developed to generate chem. fingerprints as an alternative means to the only available morphol. basis for the identification of these species. Direct anal. using real time mass spectrometry was applied to generate the chem. fingerprints of various parts viz. fruit, leaf and root of these Piper species followed by multivariate anal. for discrimination. Principal component anal. of DARTMS data served as a reliable method for species identification. Phytochem. anal. showed the presence mainly of alkaloids in various parts of Piper species. The results showed that different Piper species could be successfully differentiated using DARTMS fingerprinting together with principal component anal. and this method is promising for the rapid identification and quality control of Piper species. Metabolic engineering of Escherichia coli for biosynthesis of β-nicotinamide mononucleotide from nicotinamide was written by Liu, Yang;Yu, Bo. And the article was included in Microbial Biotechnology in 2021.SDS of cas: 1094-61-7 The following contents are mentioned in the article:

The β-NMN (NMN) is a key intermediate of an essential coenzyme for cellular redox reactions, NAD. Administration of NMN is reported to improve various symptoms, such as diabetes and age-related physiol. decline. Thus, NMN is attracting much attention as a promising nutraceutical. Here, we engineered an Escherichia coli strain to produce NMN from cheap substrate nicotinamide (NAM) and glucose. The supply of in vivo precursor phosphoribosyl pyrophosphate (PRPP) and ATP was enhanced by strengthening the metabolic flux from glucose. A nicotinamide phosphoribosyltransferase with high activity was newly screened, which is the key enzyme for converting NAM to NMN with PRPP as cofactor. Notably, the E. coli endogenous protein YgcS, which function is primarily in the uptake of sugars, was firstly proven to be beneficial for NMN production in this study. Fine-tuning regulation of ygcS gene expression in the engineered E. coli strain increased NMN production Combined with process optimization of whole-cell biocatalysts reaction, a final NMN titer of 496.2 mg l-1 was obtained. Inhibitory effects of the n-hexane, dichloromethane, ethanol and methanol extracts of the leaves of tarragon on the human carbonic anhydrase (hCA) I and II enzymes were investigated. Dichloromethane extract exhibited the strongest activity with lowest IC50 value (0.020 and 0.031 μg/mL, resp.). The chromatog. studies on the dichloromethane extract of tarragon allowed the isolation of six known strong inhibitors of hCA isoenzymes as trans-anethole, stigmasterol, herniarin, (2E,4E)-N-isobutylundeca-2,4-dien-8,10- diynamide, (2E,4E)-1-(piperidin-1-yl)undeca-2,4-diene-8,10-diyn-1-one and 1-(4′-methoxyphenyl)-1,2,3- trihydroxypropane. The compound, 1-(4′-methoxyphenyl)-1,2,3-trihydroxypropane was isolated from tarragon for the first time in this study. Among the tested compounds, 1-(4′-methoxyphenyl)-1,2,3-trihydroxypropane showed the highest inhibitiory impact on both hCA isoenzymes. The global COVID-19 epidemic has spread rapidly around the world and caused the death of more than 5 million people. It is urgent to develop effective strategies to treat COVID-19 patients. Here, we revealed that SARS-CoV-2 infection resulted in the dysregulation of genes associated with NAD+ metabolism, immune response, and cell death in mice, similar to that in COVID-19 patients. We therefore investigated the effect of treatment with NAD+ and its intermediate (NMN) and found that the pneumonia phenotypes, including excessive inflammatory cell infiltration, hemolysis, and embolization in SARS-CoV-2-infected lungs were significantly rescued. Cell death was suppressed substantially by NAD+ and NMN supplementation. More strikingly, NMN supplementation can protect 30% of aged mice infected with the lethal mouse-adapted SARS-CoV-2 from death. This report describes the development of a target-protein-oriented natural-products-isolation (TPO-NAPI) method for Hedgehog inhibitors and the direct GLI1 inhibitor, 5′-O-methyl-3-hydroxyflemingin A , which inhibited hedgehog (Hh) signal transduction and diminished characteristics of cancer stem cells. Eight natural products (including three newly described products) that directly bind to GLI1 were rapidly obtained via the TPO-NAPI method developed using GLI1 protein-immobilized beads. 5′-O-Methyl-3-hydroxyflemingin A inhibited Hh signaling (IC50 7.3 μM), leading to decreasing production of the Hh target proteins BCL2, PTCH1, and BMI1. 5′-O-Methyl-3-hydroxyflemingin A was cytotoxic to Hh-related cancer cells. CD experiments revealed that 5′-O-methyl-3-hydroxyflemingin A directly bound GLI1 (Kd = 7.7 μM). Moreover, 5′-O-methyl-3-hydroxyflemingin A diminished cancer stem cell characters of Huh7 such as sphere formation and production of the cancer stem cell marker EpCAM. These results suggest that Hh inhibitors can efficiently suppress the activity of cancer stem cells. In this study, a new type of single quadrupole mass spectrometric detector was implemented in transdermal research. The local skin pharmacokinetic properties of the plant N-alkylamides (NAAs) pellitorine and anacycline, present in an Anacyclus pyrethrum extract, and spilanthol, present in a Spilanthes acmella extract were investigated. This single quad MS detection method showed great advantages compared to the traditional UV detector. The NAAs could be identified and quantified in the samples with an ultra performance liquid chromatog. (UPLC)-single quad MS detection system, even if they were not separated, which is a requirement when using an UV-detector. Another advantage of the UPLC-MS system is that lower limit of detection values could be obtained allowing a more accurate and precise determination of the exptl. lag time in the in vitro skin permeation experiments To conclude, this single quad MS detector coupled to UPLC is a useful anal. tool with improved performance compared to high performance liquid chromatog. In X-ray crystallography, the C(=O)N center together with the three immediately adjacent atoms characteristically define a plane.Product Details of 18836-52-7

(2E,4E)-N-Isobutyldeca-2,4-dienamide (cas: 18836-52-7) belongs to amides. Amides are pervasive in nature and technology. Proteins and important plastics like Nylons, Aramid, Twaron, and Kevlar are polymers whose units are connected by amide groups (polyamides); these linkages are easily formed, confer structural rigidity, and resist hydrolysis. Amides can be freed from solvent or water by drying below their melting points. These purifications can also be used for sulfonamides and acid hydrazides.Reference of 18836-52-7

Protective effects of amide constituents from the fruit of Piper chaba on D-galactosamine/TNF-α-induced cell death in mouse hepatocytes was written by Matsuda, Hisashi;Ninomiya, Kiyofumi;Morikawa, Toshio;Yasuda, Daisuke;Yamaguchi, Itadaki;Yoshikawa, Masayuki. And the article was included in Bioorganic & Medicinal Chemistry Letters in 2008.Reference of 18836-52-7 The following contents are mentioned in the article:

The methanolic extract from the fruit of Piper chaba (Piperaceae) was found to have a hepatoprotective effect on D-galactosamine (D-GalN)/lipopolysaccharide (LPS)-induced liver injury in mice. From the Et acetate-soluble fraction, a new amide constituent named piperchabamide E (I) together with twenty known amide constituents (e.g., piperine, piperchabamides A-D, and piperanine) and two aromatic constituents were isolated as the hepatoprotective constituents. With regard to structure-activity relationships, the amide moiety and the 1,9-decadiene structure between the benzene ring and amide moiety were suggested to be important for strong inhibition of D-GalN/tumor necrosis factor-α (TNF-α)-induced death of hepatocytes. Furthermore, a principal amide constituent, piperine, dose-dependently inhibited increase in serum GPT and GOT levels at doses of 2.5-10 mg/kg (p.o.) in D-GalN/LPS-treated mice, and this inhibitory effect was suggested to depend on the reduced sensitivity of hepatocytes to TNF-α. This study involved multiple reactions and reactants, such as (2E,4E)-N-Isobutyldeca-2,4-dienamide (cas: 18836-52-7Reference of 18836-52-7).
