It is a persistent target to develop noble-metal-free catalysts that can achieve chemoselective hydrogenation of nitro compounds under mild reaction conditions. In this work, we report a new strategy for preparation of hydrogenation catalysts containing both CoNx and CoyZnS supported on N-doped porous carbon (CoNx-CoyZnS@NPC-Z). The CoNx-CoyZnS@NPC-Z catalysts were prepared by pyrolyzing the protein-metal-ion networks, followed by treatment with acid. We found that CoNxCoyZnS@NPC-3 catalyst delivers high catalytic activity and selectivity, affording almost full conversion and >98% selectivity in water/methanol mixture solvents at 90 degrees C under 5 bar of H-2 pressure for 3 h of reaction. Notably, the catalytic hydrogenation can work even under 1 bar of H-2 pressure and at room temperature with high conversion and selectivity. Besides, CoNx-CoyZnS@NPC-3 exhibits remarkable tolerance to CO or H2S poisoning and acid erosion.

A stereoselective amination/cyclization cascade process has been developed that allows for the preparation of a series of unsaturated and substituted caprolactam derivatives in good yields. This conceptually novel protocol takes advantage of the easy access and modular character of vinyl gamma-lactones that can be prepared from simple precursors. Activation of the lactone substrate in the presence of a suitable Pd precursor and newly developed phosphoramidite ligand offers a stereocontrolled ring-opening/allylic amination manifold under ambient conditions. The intermediate (E)-configured epsilon-amino acid can be cyclized using a suitable dehydrating agent in an efficient one-pot, two-step sequence. This overall highly chemo-, stereo- and regio-selective transformation streamlines the production of a wide variety of modifiable and valuable caprolactam building blocks in an operationally attractive way.

In this paper, we report on a new dielectric, a C-60-containing block polyimide (PI-b-C-60). This was realized by introducing C-60 as pendent groups into a polymer backbone. When this dielectric was used in a non-contact mode triboelectric nanogenerator (TENG), it achieved high output power and reliable operation. Compared with perfluoroalkoxy alkane film-based TENGs, the TENG based on PI-b-C-60 generated 4.3 times higher output power and a superior charge density of over 300 mu C m(-2) with a 3 times slower charge decay rate. These results are most likely due to the excellent charge-retention characteristics induced by the most negative electrostatic potential of C-60 within the backbone, and these characteristics were confirmed by surface potential measurements. Furthermore, in the course of our work, two non-contact mode applications, a keyless electronic door lock system and a speed sensor with a tone wheel for a car, were developed. Without an ion injection process being needed, very sensitive and reliable operations of the speed sensor were successfully demonstrated, even under very humid conditions (similar to 99% RH).

We report on catalysis by a fuel-induced transient state of a synthetic molecular machine. A [ 2] rotaxane molecular shuttle containing secondary ammonium/amine and thiourea stations is converted between catalytically inactive and active states by pulses of a chemical fuel (trichloroacetic acid), which is itself decomposed by the machine and/or the presence of additional base. The ON-state of the rotaxane catalyzes the reduction of a nitrostyrene by transfer hydrogenation. By varying the amount of fuel added, the lifetime of the rotaxane ON-state can be regulated and temporal control of catalysis achieved. The system can be pulsed with chemical fuel several times in succession, with each pulse activating catalysis for a time period determined by the amount of fuel added. Dissipative catalysis by synthetic molecular machines has implications for the future design of networks that feature communication and signaling between the components.

The preparation of well-dispersed graphene/polymer nanocomposites is challenging due to the poor miscibility of graphene sheets in a polymer matrix. To enhance the interaction between both phases, graphene sheets can be decorated with polymer chains. Herein, different strategies to graft poly(methyl methacrylate) (PMMA) and poly(di(ethylene glycol) ethyl ether acrylate) (PDEGA) chains at various positions on graphene oxide and reduced graphene oxide (GO/rGO) sheets are compared. Chain attachment was achieved by grafting-to and grafting-from methods. Grafting-to was performed by classical copper (I)-catalyzed alkyne azide cycloaddition. Using a grafting-from approach, PMMA and PDEGA brushes were grown from GO and rGO sheets via surface-initiated photo-induced copper-mediated polymerization (SI-photoCMP). SI-photoCMP is a robust and efficient method that allows polymerizations to be carried out under mild conditions and with reduced catalyst concentration. Moreover, the successful implementation of SI-photoCMP in a continuous-flow set-up enables easy upscaling of the system and is, therefore, a more efficient and environmentally friendly process for GO/rGO surface modification. By using the grafting-to approach, the grafting density of PMMA (M-n = 2,600 g/mol) was one chain per 990 carbons of graphene. In contrast, longer PMMA chains (M-n = 40,300 g/mol) and higher grafting density were obtained via the grafting-from method (one PMMA chain per 140 carbons of graphene).

The development of PPAR alpha/gamma dual or PPAR alpha/gamma/S pan-agonists could represent an efficacious approach for a simultaneous pharmacological intervention on carbohydrate and lipid metabolism. Two series of new phenyldiazenyl fibrate derivatives of GL479, a previously reported PPAR alpha/gamma dual agonist, were synthesized and tested. Compound 12a was identified as a PPAR pan-agonist with moderate and balanced activity on the three PPAR isoforms (alpha, gamma, delta). Moreover, docking experiments showed that 12a adopts a different binding mode in PPARy compared to PPARa or PPARS, providing a structural basis for further structure-guided design of PPAR pan-agonists. The beneficial effects of 12a were evaluated both in vitro, on the expression of PPAR target key metabolic genes, and ex vivo in two rat tissue inflammatory models. The obtained results allow considering this compound as an interesting lead for the development of a new class of PPAR pan-agonists endowed with an activation profile exploitable for therapy of metabolic syndrome.

Purine nucleotides such as ATP and ADP are important extracellular signaling molecules in almost all tissues activating various subtypes of purinoreceptors. In the brain, the P2Y(1) receptor (P2Y(1)R) subtype mediates trophic functions like differentiation and proliferation, and modulates fast synaptic transmission, both suggested to be affected in diseases of the central nervous system. Research on P2Y(1)R is limited because suitable brain-penetrating P2Y(1)R-selective tracers are not yet available. Here, we describe the first efforts to develop an F-18-labeled PET tracer based on the structure of the highly affine and selective, non-nucleotidic P2Y(1)R allosteric modulator 1-(2-[2-(tert-butyl)phenoxy]pyridin-3-yl)-3-[4- (trifluoromethoxy)phenyl]urea (7). A small series of fluorinated compounds was developed by systematic modification of the p-(trifluoromethoxy)phenyl, the urea and the 2-pyridyl subunits of the lead compound 7. Additionally, the p-(trifluoromethoxy)phenyl subunit was substituted by carborane, a boron-rich cluster with potential applicability in boron neutron capture therapy (BNCT). By functional assays, the new fluorinated derivative 1-{2-[2-(tert-butyl)phenoxy]pyridin-3-yl}-3-[4-(2-fluoroethyl) phenyl]urea (18) was identified with a high P2Y(1)R antagonistic potency (IC50 = 10 nM). Compound [F-18] 18 was radiosynthesized by using tetra-n-butyl ammonium [F-18]fluoride with high radiochemical purity, radiochemical yield and molar activities. Investigation of brain homogenates using hydrophilic interaction chromatography (HILIC) revealed [F-18]fluoride as major radiometabolite. Although [F-18]18 showed fast in vivo metabolization, the high potency and unique allosteric binding mode makes this class of compounds interesting for further optimizations and investigation of the theranostic potential as PET tracer and BNCT agent. Asymmetric synthesis based on DNA scaffolds has been actively exploited because of the advantages of DNA such as diverse tertiary structures, chemical stability, and easy handling. Since duplex DNA-based hybrid catalysts have demonstrated this remarkable capability, efforts have been made to investigate new biomolecular scaffolds. Herein, we report modular quadruplex-duplex (QD) hybrid DNA catalysts containing bipyridine ligands and hydrogen donor moieties. The conformation, thermal stability, and metal-binding ability of modified QD hybrid DNA were characterized using spectroscopy. The QD hybrid-based DNA catalysts were successfully applied to asymmetric Michael addition reactions (86% conversion and 76% ee). This study describes a new type of DNA hybrid catalyst produced by the construction of a cooperative active site with a Lewis acid and a H-bond donor.

Novel pyridine- and pyrimidine-based allosteric inhibitors are reported that achieve PDE4D subtype selectivity through recognition of a single amino acid difference on a key regulatory domain, known as UCR2, that opens and closes over the catalytic site for cAMP hydrolysis. The design and optimization of lead compounds was based on iterative analysis of X-ray crystal structures combined with metabolite identification. Selectivity for the activated, dimeric form of PDE4D provided potent memory enhancing effects in a mouse model of novel object recognition with improved tolerability and reduced vascular toxicity over earlier PDE4 inhibitors that lack subtype selectivity. The lead compound, 28 (BPN14770), has entered midstage, human phase 2 clinical trials for the treatment of Fragile X Syndrome.

The bifunctional ligand p-SCN-Bn-HOPO, which has four 1,2-hydroxypyridinone groups on a spermine backbone with an isothiocyanate linker, has been shown to be an efficient and stable chelator for Zr(iv) and, more importantly, the radioisotope Zr-89 for use in radiolabeling antibodies for positron emission tomography (PET) imaging. Previous studies of Zr-89-HOPO-trastuzumab in mice showed low background, good tumor to organ contrast, and very low bone uptake which show p-SCN-Bn-HOPO to be an important next-generation bifunctional chelator for radioimmunoPET imaging with Zr-89. However, the reported synthesis of p-SCN-Bn-HOPO involves nine steps and multiple HPLC purifications with an overall yield of about 1.4%. Herein we report an improved and efficient synthesis of p-SCN-Bn-HOPO in four steps with 14.3% overall yield which will improve its availability for further biological studies and wider application in PET imaging. The new synthetic route also allows variation in linker length and chemistries which may be helpful in modifying in vivo clearance behaviors of future agents.
