Sickle Cell Anemia (SCA) is one of the most prevalent hereditary hematological diseases worldwide. The disease is characterized by chronic inflammation, hypercoagulable state, and pro-thrombotic profile, which lead the vaso-occlusive process. In this work, we described the antiplatelet activity and the ability to reduce tumor necrosis factor-alpha (TNF-) levels of phthalimide derivatives. All compounds inhibited platelet aggregation induced by collagen and adenosine diphosphate, at levels ranging from 26.0 to 74.2% and 30.7 to 79.6%, respectively. The compounds exhibited reduced bleeding time compared to acetylsalicylic acid (ASA). Moreover, compounds 4c and 10c inhibited TNF- levels at 73.5% and 65.0%, respectively. These findings suggest that phthalimide derivatives 4c and 10c are promising lead compounds useful to prevent vaso-occlusion and inflammation associated with the sickle cell anemia. [GRAPHICS] .

Polyimides (PI) generally have a high affinity for single-walled carbon nanotubes (SWNTs), but they suffer from poor solubility in most low boiling point organic solvents and low compatibility with common resins (such as epoxy) used in composites, limiting their suitability as dispersants. PI block copolymer systems containing reactive poly(ester carbonate) s have not yet been reported and are expected to act as effective reactive dispersing agents of SWNTs. Herein, PI-derived block copolymers are synthesized via ring-opening copolymerization of lactide (LA) (a control monomer) and allyl-bearing 2-methyl-2-(allyloxycarbonyl)-propylene carbonate (MAC) from the OH-terminal ends of the PI block to produce PLA-PI-PLA (TB1, a control) and PMAC-PI-PMAC (TB2). The allyl pendant group of TB2 allows further facile functionalization to form a third series of epoxidized (EP) derivatives, i.e. PMACEP-block-PI-block-PMACEP (TB3). TB3 copolymer when mixed with a conventional structural epoxy resin forms blends that do not show inferior tensile properties compared with the epoxy, which is unusual. Furthermore, the mixing solvent tetrahydrofuran (THF) can be readily evaporated off after forming the blends. TB3-dispersed (2 wt%) SWNTs added to epoxy increased the tensile strength, modulus, and elongation at break of the resulting nanocomposite films by 40%, 34%, and 26% respectively, compared to the baseline epoxy resin. Furthermore, when TB3b triblock-dispersed SWNTs in epoxy were combined with fuzzy carbon fibers, i.e. carbon nanotube-grafted-carbon fibers (CNT-g-CF), a synergistic interfacial strength reinforcement was observed, together with shifting of the failure mode from the matrix interphase to the carbon fiber-grafted nanotube interface. The ultimate interfacial shear strength between the TB3-dispersed SWNT-epoxy matrix and the fuzzy carbon fibers (i.e., fibers having carbon nanotubes grown on them) measured via single fiber pull-out tests was 100 MPa, which was ca. 11% improvement over the baseline unsized carbon fiber in neat epoxy. To our knowledge, this is the first evidence of a synergistic enhancement in interfacial properties when fuzzy carbon fibers are combined with a SWNT-reinforced epoxy using the new epoxidized TB3 nanotube dispersing agent that forms a strong covalent TB3-epoxy interface. The new functionalizable TB3 synthesis route introduced here is generalizable to other PI-based copolymers with diverse functionalities and solvent compatibilities.

We present a novel series of radioiodinated tracers and potential theranostics for diseases accompanied by pathological function of proteins involved in choline transport. Unlike choline analogues labeled with C-11 or F-18 that are currently used in the clinic, the iodinated compounds described herein are applicable in positron emission tomography, single-photon emission computed tomography, and potentially in therapy, depending on the iodine isotope selection. Moreover, favorable half-lives of iodine isotopes result in much less challenging synthesis by isotope exchange reaction. Six of the described compounds were nanomolar ligands, and the best compound possessed an affinity 100-fold greater than that of choline. Biodistribution data of I-125-labeled ligands in human prostate carcinoma bearing (PC-3) mice revealed two compounds with a biodistribution profile superior to that of [F-18]fluorocholine.

The preparation of well-dispersed graphene/polymer nanocomposites is challenging due to the poor miscibility of graphene sheets in a polymer matrix. To enhance the interaction between both phases, graphene sheets can be decorated with polymer chains. Herein, different strategies to graft poly(methyl methacrylate) (PMMA) and poly(di(ethylene glycol) ethyl ether acrylate) (PDEGA) chains at various positions on graphene oxide and reduced graphene oxide (GO/rGO) sheets are compared. Chain attachment was achieved by grafting-to and grafting-from methods. Grafting-to was performed by classical copper (I)-catalyzed alkyne azide cycloaddition. Using a grafting-from approach, PMMA and PDEGA brushes were grown from GO and rGO sheets via surface-initiated photo-induced copper-mediated polymerization (SI-photoCMP). SI-photoCMP is a robust and efficient method that allows polymerizations to be carried out under mild conditions and with reduced catalyst concentration. Moreover, the successful implementation of SI-photoCMP in a continuous-flow set-up enables easy upscaling of the system and is, therefore, a more efficient and environmentally friendly process for GO/rGO surface modification. By using the grafting-to approach, the grafting density of PMMA (M-n = 2,600 g/mol) was one chain per 990 carbons of graphene. In contrast, longer PMMA chains (M-n = 40,300 g/mol) and higher grafting density were obtained via the grafting-from method (one PMMA chain per 140 carbons of graphene).

Peroxidase-generated proximity labeling is in widespread use to study subcellular proteomes and the protein interaction networks in living cells, but the development of subcellular RNA labeling is limited. APEX-seq has emerged as a new method to study subcellular RNA in living cells, but the labeling of RNA still has room to improve. In this work, we describe 4-thiouridine (s(4)U)-enhanced peroxidase-generated biotinylation of RNA with high efficiency. The incorporation of s(4)U could introduce additional sites for RNA labeling, enhanced biotinylation was observed on monomer, model oligo RNA and total RNA. Through the s(4)U metabolic approach, thein vivoRNA biotinylation efficiency by peroxidase catalysis was also dramatically increased, which will benefit RNA isolation and study for the spatial transcriptome.

Covalent agonists of PPAR cause unique receptor conformational changes and behave as selective PPAR modulators, whereas there are few covalent agonists other than endogenous unsaturated fatty acids metabolites. Previously, we established a cell-based strategy to identify new PPAR ligands and synthesized a new-type of covalent agonist that possesses the hybrid structure of a plant-derived cinnamic acid derivative and GW9662, a covalent antagonist. Herein, we report six analogues that differ in how the two fragments are linked together. Compounds with a simplified linker showed potent agonistic activity with improved EC50 values (less than 5 nM), indicating that close proximity between the two fragments improves binding affinity. When the position of cinnamic acid moiety was placed at 4 carbon of aniline ring, PPAR agonist activity was completely abolished. Docking studies suggested that the activation profile likely depends on interaction with the cavity around helix 3, -sheet, and -loop region in the ligand-binding domain. Furthermore, a cell-based assay revealed that agonist-type compounds activate PPAR transcription in a manner dependent on covalent linkage with the Cys285 residue leading to prolonged transactivation. This activation feature reflects pharmacological benefits of covalent drugs, suggesting that these hybrid compounds may serve as potential leads for a new-class of covalent PPAR ligands.

A collection of potent antimicrobials consisting of novel 1,3-bis-benzoic acid and trifluoromethyl phenyl derived pyrazoles has been synthesized and tested for antibacterial activity. The majority of trifluoromethyl phenyl derivatives are highly potent growth inhibitors of Gram-positive bacteria and showed low toxicity to human cultured cells. In particular, two compounds (59 and 74) were selected for additional studies. These compounds are highly effective against Staphylococcus aureus as shown by a low minimum inhibitory concentration (MIC), a bactericidal effect in time-kill assays, moderate inhibition of biofilm formation as well as biofilm destruction, and a bactericidal effect against stationary phase cells representing non-growing persister cells. Multistep resistance assays showed a very low tendency for S. aureus and Enterococcus faecalis to develop resistance through mutation. Additionally, in vivo mouse model studies showed no harmful effects at doses up to 50 mg/kg using 14 blood plasma organ toxicity markers or TUNEL assay in liver and kidney. Investigations into the mode of action by performing macromolecular synthesis inhibition studies showed a broad range of inhibitory effects, suggesting targets that have a global effect on bacterial cell function. In this paper, we report on a new dielectric, a C-60-containing block polyimide (PI-b-C-60). This was realized by introducing C-60 as pendent groups into a polymer backbone. When this dielectric was used in a non-contact mode triboelectric nanogenerator (TENG), it achieved high output power and reliable operation. Compared with perfluoroalkoxy alkane film-based TENGs, the TENG based on PI-b-C-60 generated 4.3 times higher output power and a superior charge density of over 300 mu C m(-2) with a 3 times slower charge decay rate. These results are most likely due to the excellent charge-retention characteristics induced by the most negative electrostatic potential of C-60 within the backbone, and these characteristics were confirmed by surface potential measurements. Furthermore, in the course of our work, two non-contact mode applications, a keyless electronic door lock system and a speed sensor with a tone wheel for a car, were developed. Without an ion injection process being needed, very sensitive and reliable operations of the speed sensor were successfully demonstrated, even under very humid conditions (similar to 99% RH).

Sustainable development demands an environmentally friendly and efficient method for the hydrogenation of organic molecules, including the hydrogenation of functionalized nitroarenes. In this study, a highly active and selective metal-organic framework-supported palladium catalyst was prepared for the catalytic hydrogenation of nitroarenes. High selectivity (>99%) and excellent yield (98%) of aniline were realized after 2 hours in ethanol under hydrogen (1 atm) at room temperature. The reductions were successfully carried out in the presence of a wide range of other reducible functional groups. More importantly, the catalyst was very stable without the loss of its catalytic activity after five cycles.
