Enhanced Performance of Reagent-Less Carbon Nanodots Based Enzyme Electrochemical Biosensors

This work reports on the advantages of using carbon nanodots (CNDs) in the development of reagent-less oxidoreductase-based biosensors. Biosensor responses are based on the detection of H2O2, generated in the enzymatic reaction, at 0.4 V. A simple and fast method, consisting of direct adsorption of the bioconjugate, formed by mixing lactate oxidase, glucose oxidase, or uricase with CNDs, is employed to develop the nanostructured biosensors. Peripherical amide groups enriched CNDs are prepared from ethyleneglycol bis-(2-aminoethyl ether)-N,N,N’,N’-tetraacetic acid and tris(hydroxymethyl)aminomethane, and used as precursors. The bioconjugate formed between lactate oxidase and CNDs was chosen as a case study to determine the analytical parameters of the resulting L-lactate biosensor. A linear concentration range of 3.0 to 500 mu M, a sensitivity of 4.98 x 10(-3) mu A.mu M-1, and a detection limit of 0.9 mu M were obtained for the L-lactate biosensing platform. The reproducibility of the biosensor was found to be 8.6%. The biosensor was applied to the L-lactate quantification in a commercial human serum sample. The standard addition method was employed. L-lactate concentration in the serum extract of 0.9 +/- 0.3 mM (n = 3) was calculated. The result agrees well with the one obtained in 0.9 +/- 0.2 mM, using a commercial spectrophotometric enzymatic kit.

Solid-liquid equilibria and excess enthalpies in binary mixtures of acetophenone with some aliphatic amides

Solid-liquid equilibria for the binary systems of acetophenone and {N-methylformamide, or N,N-dimethylformamide, or N,N-dimethylacetamide, or N-methyl-2-pyrrolidone} were determined by the cloudpoint and DSC techniques. Coadministration of chemotherapy and PI3K/Akt pathway treatment with multistage acidity/CathB enzyme-responsive nanocarriers for inhibiting the metastasis of breast cancer

As the principal reason for the inducement of high mortality, tumor metastasis is regulated by different pathways owing to its complexity and multistep process. In order to inhibit the proliferation and metastasis of human breast cancer simultaneously, controlling the codelivery of chemotherapeutics and pathway inhibitors precisely has been considered as a high-potential strategy to accurately eliminate tumor metastasis. In this study, polymer PLGA-p-PEI-DA was synthesised and automatically assembled into a cascade trinity response drug delivery system, i.e., PPP-DA/NPs (PLGA: poly(lactin-co-glycolic acid), PEI: polyethyleneimine, DA: 2,3-dimethylmaleic anhydride). In the tumor microenvironment, PPP-DA/NPs could remove the outer DA molecules via the pH-sensitive hydrolysis of beta-carboxylic amide bonded with DA and PEI. Then, PPP-DA/NPs were broken up owing to the enzymatically cleavable GFLGF (Gly-Phe-Leu-Gly-Phe) linker. The structure of the polymer and the properties of PPP-DA/NPs were evaluated in detail. Moreover, studies on the antitumor metastasis efficiency and antitumor mechanism of PPP-DA/NPs were carried out in detail. As demonstrated in this study, PPP-DA/NPs could reverse the potential in pH 6.8 PBS and showed elevated cellular uptake efficiency. Moreover, PPP-DA/NPs exhibited strong antitumor metastasis ability in vitro and in vivo. The tumor inhibiting rate (TIR) of PPP-DA/NPs (68.4%) was significantly higher than that of docetaxel (DTX) (5.9%). The antitumor mechanistic studies confirmed that PPP-DA/NPs could down-regulate the expressions of Akt, MMP-9 and pro-caspase-3/9 protein, as indicated by western blot analysis. This multifunctional drug delivery system (DDS) is highly selective and effective in inhibiting tumor metastasis, which shows a great potential in inventing smart nanocarriers for targeted tumor-metastasis therapy.

Human mass balance study and metabolite profiling of C-14-niraparib, a novel poly(ADP-Ribose) polymerase (PARP)-1 and PARP-2 inhibitor, in patients with advanced cancer

Niraparib is an investigational oral, once daily, selective poly(ADP-Ribose) polymerase (PARP)-1 and PARP-2 inhibitor. In the pivotal Phase 3 NOVA/ENGOT/OV16 study, niraparib met its primary endpoint of improving progression-free survival (PFS) for adult patients with recurrent, platinum sensitive, ovarian, fallopian tube, or primary peritoneal cancer in complete or partial response to platinum-based chemotherapy. Significant improvements in PFS were seen in all patient cohorts regardless of biomarker status. This study evaluates the absorption, metabolism and excretion (AME) of C-14-niraparib, administered to six patients as a single oral dose of 300 mg with a radioactivity of 100 mu Ci. Total radioactivity (TRA) in whole blood, plasma, urine and faeces was measured using liquid scintillation counting (LSC) to obtain the mass balance of niraparib. Moreover, metabolite profiling was performed on selected plasma, urine and faeces samples using liquid chromatography – tandem mass spectrometry (LC-MS/MS) coupled to off-line LSC. Mean TRA recovered over 504 h was 47.5% in urine and 38.8% in faeces, indicating that both renal and hepatic pathways are comparably involved in excretion of niraparib and its metabolites. The elimination of C-14-radioactivity was slow, with t(1/2) in plasma on average 92.5 h. Oral absorption of C-14-niraparib was rapid, with niraparib concentrations peaking at 2.49 h, and reaching a mean maximum concentration of 540 ng/mL. Two major metabolites were found: the known metabolite M1 (amide hydrolysed niraparib) and the glucuronide of M1. Based on this study it was shown that niraparib undergoes hydrolytic, and conjugative metabolic conversions, with the oxidative pathway being minimal.

Polyacrylonitrile modified partially reduced graphene oxide composites for the extraction of Hg(II) ions from polluted water

Polyacrylonitrile nanoparticles grafted on ethylene diamine functionalized partially reduced graphene oxide (PAN-PRGO) was prepared via in situ emulsion polymerization and was further modified to contain amidoxime, amdinoethylene diamine, and carboxylic groups on the surface of the graphene nanosheets via partial hydrolysis of the nitrile groups on the polymer chains of the composite using (4% NaOH, 20 min) (HPAN-PRGO). The properties and morphologies of the prepared composites were compared through FTIR, UV-Vis, Raman spectra, XRD, SEM, TEM, and XPS analysis. The results revealed that polyacrylonitrile nanoparticles were grafted on the surface of the aminated graphene oxide nanosheets via the reaction between the free amino groups of the ethylene diamine modified graphene oxide nanosheets and nitrile groups of acylonitrile (AN). The obtained HPAN-PRGO composite was evaluated for its chelating property with Hg(II) ions. The effect of initial pH, initial concentration of the Hg(II), adsorbent dose, and contact time on the extraction of Hg(II) ions using HPAN-PRGO were investigated. The adsorption experiments indicated that HPAN-PRGO exhibits higher affinity toward Hg(II). The maximum uptake capacity for the extraction of Hg(II) ions on HPAN-PRGO was 324.0 mg/g at pH 5. The HPAN-PRGO shows a 100% removal of Hg(II) at concentrations up to 50 ppm, and the adsorption is exceptionally rapid showing more than 80.0% removal within 15 min and 100.0% of q(e) within 1.5 h at 800 ppm concentration. The Langmuir isotherm model and pseudo-second-order kinetic model have showed good fitness with the practical data. The XPS analysis of HPAN-PRGO before and after adsorption revealed the chelation adsorption mechanism between mercury and amine, amide, amidoxime, and carboxylic groups. After six adsorption-desorption cycles, the HPAN-PRGO could retain more than 90.0% of its original adsorption capacity. These results confirmed that HPAN-PRGO has exceptional performance for the removal of Hg(II) from wastewater. [GRAPHICS] .
