The amidation of long-chain fatty acids is the key step for preparing surfactants with excellent interfacial activity. Gas chromatography-mass spectrometry was employed to detect the reactants and products in the direct amidation reactions. The conversion and the concentration of the amides in the reaction process were also investigated to determine the best catalyst, the reaction rate constants, and activation energy. Tyrosine-containing cyclic dipeptides based on a diketopiperazine (DKP) ring are studied under jet-cooled conditions using resonance-enhanced multi-photon ionisation (REMPI), conformer-selective IR-UV double resonance vibrational spectroscopy and quantum chemical calculations. The conformational landscape of the dipeptide containing natural L tyrosine (Tyr), namely c-LTyr-LTyr strongly differs from that of its diastereomer c-LTyr-DTyr. Weak NH and CH interactions are observed, which are slightly different in c-LTyr-LTyr and c-LTyr-DTyr. These structures are identical to those of LL and LD cyclo diphenylalanine, which only differ from c-Tyr-Tyr by the absence of hydroxyl on the benzene rings. While this is the only conformation observed for c-LTyr-DTyr, c-LTyr-LTyr exhibits an additional form stabilised by the interaction of the two hydroxyls, in which the two aromatic rings are in a stacked geometry. Stereochemical effects are still visible in the radical cation, for which one structure is observed for c-LTyr-DTyr, while the spectrum of the c-LTyr-LTyr radical cation is explained in terms of two co-existing structures.

The review reports an aqueous solution behavior of commercially available and easy-to-use polymers, i.e., Pluronics (R)- and PNIPAM-based block copolymers. Both the polymers are stimuli responsive in nature. The present review covers the different aspects of aggregation behavior of Pluronics (R)- and PNIPAM-based block copolymeric micelles. Here, a comparison of physical properties such as EO-PO block, CP, CMC and CMT is made. Such physical parameters can be modulated with ease by the presence of external stimuli, viz. electrolytes, organic additives such as alcohols, phenols, amides and acids, different types of surfactants and water-soluble polymers, and also by modification of end groups of Pluronics (R). But in this review, our main focus is to study the addition of salts and non-electrolytes on the aggregation behavior of Pluronics (R). With the help of above parameters, users can get idea about the stability, partition coefficient, solubilization capacity, etc. In analogy with Pluronics (R), PNIPAM is also a thermo-responsive polymer with similar to 32 degrees C lower critical solution temperature (LCST). However, the LCST is independent of the degree of polymerization, i.e., molecular weight of homopolymer. However, the LCST can be tuned in the presence of external stimulus. PNIPAM-based di-block copolymers form various morphologies in different environments. An inverted morphology by double-hydrophilic-block copolymers is also possible. According to US and British Pharmacopoeia, some of the Pluronics (R) and PNIPAM are recognized as pharmaceutical excipients. Therefore, they have been extensively used for various pharmaceutical formulations. The other applications of these polymers are tissue engineering, bioseparation devices, active membranes, biosensors, rheological modifier, lithium batteries, etc. [GRAPHICS] .

SpyTag is a peptide that forms a spontaneous amide bond with its protein partner SpyCatcher. Dehydrogenative amide bond formation from alcohols and amines has been regarded as an atom-economic and sustainable process. Among various catalytic systems, N-heterocyclic carbene (NHC)-based Ru catalytic systems have attracted growing interest due to the outstanding properties of NHCs as ligands. Herein, an NHC/Ru complex (1) was prepared and its structure was further confirmed with X-ray crystallography. In the presence of Cs2CO3, two NHC/Ru-based catalytic systems were disclosed to be active for this amide synthesis. System A, which did not contain any added ligand, required a catalyst loading of 1.00 mol%. Interestingly, improved catalytic performance was realized by the addition of an NHC precursor (L). Optimization of the amounts of L and other conditions gave rise to system B, a much more potent system with the Ru loading as low as 0.25 mol%. Moreover, an NHC-Ru-carbonate complex 6 was identified from the refluxing toluene of 1 and Cs2CO3, and further investigations revealed that 6 was an important intermediate for this catalytic reaction. Based on the above results, we claimed that the role of Cs2CO3 was to facilitate the formation of key intermediate 6. On the other hand, it provided the optimized basicity for the selective amide formation.

In contrast, [Y(BIPM)(I)(THF)(2)] reacted with the more sterically demanding potassium bis(silyl)amides KN*(dagger) (N*(dagger) = {N((SiMe2Bu)-Bu-t)((SiPr3)-Pr-i)}) and KN dagger dagger (N-dagger dagger = {N((SiPr3)-Pr-i)(2)}) to afford the methanide cyclometalate complexes [Y(H-BIPM){N((SiPr3)-Pr-i)((SiBuMeCH2)-Bu-t)-kappa(2)-N,C)}] (4) and [Y(H-BIPM){N((SiPr3)-Pr-i)[(SiPr2)-Pr-i(CHMeCH2)]-kappa(2)-N,C}] (5), respectively. Preeclampsia (PE) is a hypertensive disorder of pregnancy and one of the leading contributors to both maternal and perinatal morbidity and mortality. Reliable diagnostic parameters unique to the disorder that accurately define and diagnose PE are currently unavailable. Here, we evaluate the capability of an amyloid-targeting aryl cyano amide (ARCAM-1) fluorophore to identify PE patients from analysis of urine samples. Our results reveal that this probe can distinguish patients with PE from gestationally healthy patients and patients suffering from non-PE hypertension, highlighting the potential for amyloid-targeting fluorophores to help identify PE patients during pregnancy.

Two main amyloid-beta peptides of different length (A beta(40) and A beta(42)) are involved in Alzheimer’s disease. Their relative abundance is decisive for the severity of the disease and mixed oligomers may contribute to the toxic species. However, little is know about the extent of mixing. To study whether A beta(40) and A beta(42) co-aggregate, we used Fourier transform infrared spectroscopy in combination with C-13-labeling and spectrum calculation and focused on the amide I vibration, which is sensitive to backbone structure. Mixtures of monomeric labeled A beta(40) and unlabeled A beta(42) (and vice versa) were co-incubated for similar to 20 min and their infrared spectrum recorded. The position of the main C-13-amide I’ band shifted to higher wavenumbers with increasing admixture of C-12-peptide due to the presence of C-12-amides in the vicinity of C-13-amides. The results indicate that A beta(40) and A beta(42) form mixed oligomers with a largely random distribution of A beta(40) and A beta(42) strands in their beta-sheets. The structures of the mixed oligomers are intermediate between those of the pure oligomers. There is no indication that one of the peptides forces the backbone structure of its oligomers on the other peptide when they are mixed as monomers. We also demonstrate that isotope-edited infrared spectroscopy can distinguish aggregation modulators that integrate into the backbone structure of their interaction partner from those that do not. As an example for the latter case, the pro-inflammatory calcium binding protein S100A9 is shown not to incorporate into the b-sheets of A beta(42).
