The present study focuses on novel degradation products of alectinib hydrochloride generated from forced degradation study. In this process, an analytical method was developed that is not only compatible with liquid chromatography-mass spectrometry but sufficiently efficient to capture each of the possible degradation products’ peaks with effective separation from alectinib peak. Alectinib hydrochloride was exposed to acidic, alkali, oxidation, photolytic, and thermal conditions as recommended by the International Conference on Harmonization guidelines. No major degradation was observed under acidic, alkali, photolytic and thermal conditions however the drug was observed to be significantly degraded under oxidative stress conditions generating four novel degradation products that are not reported in the literature. The degradation products were isolated using preparative high-performance liquid chromatography and then structurally elucidated by Fourier transform infrared spectroscopy, nuclear magnetic resonance spectroscopy, and liquid chromatography-mass spectrometry studies. Most importantly, three degradation products were identified as constitutional isomers. The probable degradation products were identified as N-oxide impurity, epoxide impurity, N-hydroxy impurity, and amide impurity, respectively. Moreover, pathway of formation of these degradation products was also postulated in detail.

Although the reversibility of 10,12-pentacosadiynoic amino meta-acid (PCDA-mBzA) against temperature and pH was reported, the modulation of reversibility by ion adsorption at terminal functional group has not been investigated. In this work, we developed a simple method for modulating the reversibility of PCDA-mBzA films upon a thermal stimulus by cadmium ion adsorption inducing the breakage of the outer hydrogen bonding of two hydrogen bonds, which are responsible for the reversible properties of PCDA-mBzA. External reflection-Fourier transform infrared (ER-FTIR) analyses revealed that the hydrogen bonding between the carboxylic acid groups was broken through ion adsorption and only a single hydrogen bond between the amide groups remained in the PCDA-mBzA polymer. In addition, PCDA-mBzA films could recover their original property through cadmium ion desorption. These results present that the transition between reversibility and irreversibility can be modulated artificially simply through the adsorption and desorption of metal ions. The purpose of this study was to investigate the potential of immobilized lead- and cadmium-resistant Pseudomonas putida strain PT to remove heavy metals from aqueous medium under extreme conditions. The tolerance and accumulation of cadmium and lead ions by strain PT were investigated by minimal inhibitory concentration (MIC) determination and polymerase chain reaction (PCR) of cadA gene, respectively. The surface chemical functional groups of P. putida PT involved in the metal biosorption were identified by Fourier transform infrared (FTIR). Pseudomonas putida PT was immobilized in three matrices include carboxy-methyl cellulose (CMC), rice bran, and a new composite made of alginate, polyvinyl alcohol (PVA), and CaCO3 to prepare heavy metal adsorbent. The biosorbents were analyzed by SEM, and their metal removal capability was assayed in two consecutive cycles by atomic absorption spectroscopy. The viability of immobilized bacterial cells was determined by flow cytometry during storage at 4 degrees C and exposure to the environmental stresses (pH and temperature). The results showed that PT strain was resistant up to 10mM Pb2+ and 8mM Cd2+. FTIR analysis revealed that alcohol, sulfur, phosphate, esters, and amide groups played important roles in metal biosorption process and, also change in metabolic reactions like hydration and polyesters accumulation was observed after metal biosorption. The presence of cadA gene, a heavy metal translocating pump-coding gene, indicated the ability of metals bioaccumulation by the PT strain. Immobilized cells in alginate-PVA-CaCO3 and rice bran showed the highest metal removal efficiency for Pb2+ as 75% and Cd2+ as 96.7%, respectively. Metal adsorbents were reusable, and the highest removal efficiency in the second cycle was observed in inoculated alginate-PVA-CaCO3 (79.5% Pb2+ and 45% Cd2+). Flow cytometric analysis represented that the immobilized cell viability was retained (<97%) after 4weeks storage at 4 degrees C. Viability under two environmental stresses in all matrices was as follows: <96% at 25 degrees C, <87% at 45 degrees C, <85% at pH4,<96% at pH7, and<89% at pH11. The results signify that these metal adsorbents are efficient technological tools for bioremediation even in harsh environmental conditions. Ultrafast 2D-IR spectroscopy is a powerful tool for understanding the spectroscopy and dynamics of biological molecules in the solution phase. A number of recent studies have begun to explore the utility of the information-rich 2D-IR spectra for analytical applications. Here, we report the application of ultrafast 2D-IR spectroscopy for the detection and classification of bacterial spores. 2D-IR spectra of Bacillus atrophaeus and Bacillus thuringiensis spores as dry films on CaF2 windows were obtained. The sporulated nature of the bacteria was confirmed using 2D-IR diagonal and off-diagonal peaks arising from the calcium dipicolinate CaDP3H(2)O biomarker for sporulation. Distinctive peaks, in the protein amide I region of the spectrum were used to differentiate the two types of spore. The identified marker modes demonstrate the potential for the use of 2D-IR methods as a direct means of spore classification. We discuss these new results in perspective with the current state of analytical 2D-IR measurements, showing that the potential exists to apply 2D-IR spectroscopy to detect the spores on surfaces and in suspensions as well as in dry films. The properties of gelatin oven dried at three different temperatures from unicorn leatherjacket (L50, L60 and L70) and reef cod skin (R50, R60 and R70) were investigated. Differential scanning calorimetry (DSC) showed a higher melting temperature in unicorn leatherjacket skin gelatin (115.54-120.80 degrees C) than reef cod skin (106.25-118.92 degrees C). Foaming capacity and stability and emulsion stability were higher in unicorn leatherjacket skin gelatin. Emulsion activity index, fat binding and water holding capacities were higher in reef cod skin gelatin. Different oven drying temperatures has not affect the yield and quality of gelatin. This study could pave the way to utilize oven drying for the extraction of gelatin from fish skin.

The present work describes a highly efficient synthetic strategy for amides via oxidative coupling of benzaldehydes or benzylamines with N-substituted formamides using a heterogeneous Co/Al hydrotalcite-derived catalyst in the presence of TBHP. A series of Co/Al hydrotalcite-derived catalysts (Cat-2, Cat-3, and Cat-4 with the Co2+/Al3+ molar ratio in the synthesis mixture as 1/1, 2/1 and 3/1) have been prepared by a simple co-precipitation method and characterized using powder XRD, XPS, FEG-SEM, EDS, FT-IR, DTG-TGA and N-2 physical adsorption techniques. Among the as-prepared catalysts, Cat-3 exhibited excellent catalytic activity towards the direct amidation of benzaldehydes as well as benzylamines bearing various substituents into the corresponding amides at 100 degrees C using TBHP as an oxidant. The mechanistic investigation of the amidation reaction revealed that the reaction follows a radical pathway. Furthermore, the catalyst is easily separable and recyclable without considerable loss in catalytic activity.
