Antioxidant functionalization of biomaterials via reactive extrusion

Concern over environmental contamination and the need for industrially produced functional biopolymers motivates the utilization of reactive extrusion for functional grafting. Antioxidant nonmigratory active materials were synthesized through 1,3-bis(4,5-dihydro-2-oxazolyl)benzene ring opening polymerization with bio-based poly(lactic acid) (PLA) and antioxidant nitrilotriacetic acid (NTA). Grafting was confirmed through the introduction of new alkyl stretching, disappearance of the characteristic 1,3-bis(4,5-dihydro-2-oxazolyl)benzene band, and emergence of new amide band on attenuated total reflectance Fourier transform infrared spectroscopy. X-ray photoelectron spectroscopy (XPS) showed stepwise increases in atomic nitrogen percentage and shifts in proportion of CC:CO bonding, confirming grafting and surface orientation. Antioxidant samples exhibited significant increases in carboxylate density, ranging from 0.75 +/- 0.04 to 3.11 +/- 0.04 nmol/cm(2). Functionalized films demonstrated significant antioxidant properties with Trolox((eq)) ranging from 0.36 +/- 0.02 to 0.89 +/- 0.07 nmol/cm(2) according to radical scavenging studies and delayed greater than 52% of ascorbic acid degradation. This work develops a rapid functionalization method for biopolymers and displays efficacy in producing sustainable nonmigratory active materials.

Interfacial Effect of Hydration Structures of Hydroxyapatite Nanoparticle Films on Protein Adsorption and Cell Adhesion States

The synthesized elliptical hydroxyapatite (E-HAp) and needle-like HAp (N-HAp) nanoparticles (NPs) were electrophoretically deposited on a gold (Au) substrate. A comparative study of the hydration layers on E-HAp, N-HAp, and Au films was achieved to investigate the interfacial effect of the hydration layers on the conformation of the adsorbed fibrinogen (Fgn) and fibroblast adhesion properties. As a result, the ratios of three types of hydration layer states (free water, intermediate water, nonfreezing water) analyzed by a Fourier transform infrared (FT-IR) spectral deconvolution of the O-H stretching absorption band were investigated. The ratio of the bonding water state (i.e., intermediate and nonfreezing water molecules) is almost the same between two HAp films, and the E-HAp film with an elliptical shape and smaller particle size exhibited the smallest ratio of nonfreezing water, which can suppress the denaturation of the adsorbed protein. Subsequently, FT-IR spectral deconvolution results of the amide I band of the adsorbed Fgn on the E-HAp film indicated the higher proportion of a-helix and beta-sheet structures as compared with those on the N-HAp and Au films, suggesting that the smaller proportion of nonfreezing waters would play a significant role in the stereoscopic Fgn conformation. In the culture of fibroblasts, FT-IR spectra of the adhered cells on the E-HAp, N-HAp, and Au films exhibited different absorbance intensities of the amide A, I, II, and III bands, suggesting a different amount of collagen-producing states by the cells, which were also supported by immunostaining results of the collagen type I. Therefore, the different hydration structures on the films clearly influenced the conformation of the adsorbed protein, and the preferential conformation was found at the interfaces between the fibroblasts and the underground E-HAp films.

We show that the most efficient protocol is achieved by the CCA-catalyzed pyrophosphorolysis removal of the natural A76 in equilibrium with the addition of the appropriate ATP analog to synthesize the modified 3′-end. This protocol for 3’-amino-tailing affords quantitative and stable attachment of a broad range of amino acids to tRNA, indicating its general utility for studies of aminoacyl-tRNAs in both canonical and noncanonical activities.

(Nano)Fibrillar morphology development in biobased poly(butylene succinate-co-adipate)/poly(amide-11) blown films

Thin films with (nano)fibrillar morphologies were successfully obtained in fully-biobased poly(butylene succinate-co-adipate)/poly(amide-11) blends (PBSA/PA11, 85/15 wt/wt) using an extrusion-blowing process. Impacts of PA11 grade and take-up ratio on the morphology of PBSA/PA11 were particularly highlighted. Scanning electron microscopy analyses indicated that PA11 with high melt volume-flow rates are beneficial to the development of (nano)fibrillar morphologies in PBSA/PA11 blown film. On the contrary, unstable film blowing processing without fibrillar morphologies was attested for PA11 with low melt volume-flow rates. Increasing the take-up ratio during extrusion-blowing of PBSA/PA11 clearly generates finer PA11 (nano)fibrils into PBSA. Fibril diameters down to 300 nm could be reached with an optimal PA11 grade promoting enhanced mechanical properties (higher ductility and toughness). The formation of stable PA11 (nano)fibrils into PBSA is discussed via rheological assessments of viscosity/elasticity ratio. A specific attention was finally paid to the PBSA strain-hardening behavior in PBSA/PA11 using elongational rheological tests. PA11 (nano)fibrillation helps maintaining the strong PBSA strain-hardening and thus play a major role on the processability of PBSA/PA11 blends by extrusion blowing. As a conclusion, the PA11 grade represents a crucial parameter to control the production of PBSA/PA11 blown films with refined (nano)fibrillar structures and enhanced physico-chemical properties.

Analysis of tuberculosis disease through Raman spectroscopy and machine learning

We present the effectiveness of Raman spectroscopy (RS) in combination with machine learning for screening and analysis of blood sera collected from tuberculosis patients. Blood samples of 60 patients have confirmed active pulmonary tuberculosis and 14 samples of healthy age matched control were used in the current study. Mechanically Robust and Reprocessable Acrylate Vitrimers with Hydrogen-Bond-Integrated Networks for Photo-3D Printing

Reprocessable acrylate vitrimer needs to enhance its strength to expand the application in photo-three-dimensional (photo-3D) printing. However, the methods for improving mechanical properties by the addition of nanofillers or a multifunctional resin into acrylate vitrimers are inappropriate for photo-3D printing due to the low curing speed of photopolymerization induced by weakening light transmittance or reduction of dimensional accuracy caused by large shrinkage. At present, we demonstrate a new strategy for developing a kind of mechanically robust and reprocessable 3D printing thermosets by combining hydrogen bonds and exchangeable beta-hydroxyl esters into acrylate vitrimers. To realize this purpose, diacrylate prepolymer containing beta-hydroxyl esters was first synthesized from glycidyl methacrylate and suberic acid. Then, the resin formulations for 3D printing comprising the synthesized diacrylate prepolymer together with acrylamide generate exchanged beta-hydroxyl ester and pendent amide in cross-linked networks. Here, hydrogen bonds resulting from the amide group as sacrificial bonds dissipate vast mechanical energy under an external load. With the inclusion of 20 wt % acrylamide, the average tensile strength and Young’s modulus are up to 40.1 and 871 MPa, which increased by about 4.4 and 3.85 times, respectively. The network rearrangement of cross-linked vitrimers can be achieved through the dynamic ester exchange reactions with gradual disappearance of hydrogen bonds at elevated temperatures, imparting reprocessability into the printed structures. Various photo-3D printing or UV irradiation shapes were successfully produced, and these dissolved in ethylene glycol could be remolded again.
