Response of microbes to biochar strengthen nitrogen removal in subsurface flow constructed wetlands: Microbial community structure and metabolite characteristics

Four subsurface flow constructed wetlands (SFCWs) were constructed on the basis of the volume ratio of biochar in common gravel (0%, 10%, 20%, and 30%) for the evaluation of microbe and metabolite characteristics response to biochar addition. The results showed that the biochar added SFCWs provided higher removal efficiencies for ammonium (49.69%-63.51%) and total nitrogen (81.83%-86.36%), compared with pure gravel packed SFCWs for ammonium (47.40%) and total nitrogen (80.75%), respectively. Illumina MiSeq sequencing results revealed that the dominant phyla were Proteobacteria, Bacteroidetes, and Firmicutes. Biochar addition can improve the removal of nitrogen by altering microbial community and increasing the relative abundance of Thauera, Candidatus Competibacter, Dechloromonas, Desulfobulbus, Chlorobium, and Thiobacillus. Protein and humic substances were the primary components of extracellular polymeric substance (EPS) in SFCWs. The amount of total EPS considerably decreased with biochar addition, which caused a shift in the EPS functional groups including carbonyl of protein, amide, and hydroxyl groups. Moreover, biochar could enhance the high molecular weight compounds metabolized into low molecular compounds. The endocannabinoid (eCB) system is exquisitely sensitive to these hormonal changes and is a significant regulator of emotional behavior. The impact of peripheral inflammation on central eCB function, and whether this is related to the development of these behavioral comorbidities remains to be determined. To examine this, we employed the trinitrobenzene sulfonic acid-induced model of colonic inflammation (colitis) in adult, male, Sprague Dawley rats to produce sustained peripheral inflammation. Colitis produced increases in behavioral measures of anxiety and elevations in circulating corticosterone. These alterations were accompanied by elevated hydrolytic activity of the enzyme fatty acid amide hydrolase (FAAH), which hydrolyzes the eCB anandamide (AEA), throughout multiple corticolimbic brain regions. This elevation of FAAH activity was associated with broad reductions in the content of AEA, whose decline was driven by central corticotropin releasing factor type 1 receptor signaling. Colitis-induced anxiety was reversed following acute central inhibition of FAAH, suggesting that the reductions in AEA produced by colitis contributed to the generation of anxiety. These data provide a novel perspective for the pharmacological management of psychiatric comorbidities of chronic inflammatory conditions through modulation of eCB signaling.

Meat quality and Raman spectroscopic characterization of Korat hybrid chicken obtained from various rearing periods

Meat quality attributes vary with chicken age. Understanding the relationship between poultry age and the quality of the meat would be beneficial for efficient poultry farming to meet market needs. The Korat hybrid chicken (KC) is a new crossbred chicken whose meat quality is distinct from that of commercial broiler (CB) chickens and has not been well characterized. In this study, we characterized the physico-chemical properties of KC meat and correlate the findings with Raman spectral data. The protein content of KC breast and thigh meat increased with age. The pH of thigh meat decreased, while the water-holding capacity of breast meat increased as the age of the chickens increased. The amount of cholesterol in breast meat decreased as the rearing period was extended. Inosine 5′-monophosphate and guanosine 5′-mono phosphate of breast meat decreased as KC grew older. The shear force values of meat from older birds increased concomitantly with an increase in total collagen. Principle component analysis revealed that the meat quality of CB was greatly different from that of KC meat. High shear force values of KC meat at 20 wk of age were well correlated with an increase in the beta-sheet structure (amide I) and amide III of collagen. Raman spectra at 3,207 cm(-1) and relative alpha-helical content were negatively correlated with shear force values of KC breast meat. These could be used as markers to evaluate KC meat quality.

Green extraction of perilla volatile organic compounds by pervaporation

Volatile organic compounds (VOCs) present in perilla essential oil are of high interest in medicinal and food processing. In this work, pervaporation was implemented to extract the valuable perilla VOCs from dilute aqueous solutions as a green process. Three representative VOCs of perilla (i.e., limonene, linalool, and perillaldehyde) having different functional groups were selected as model components, and poly(ether-block-amide) (PEBA) and polydimethylsiloxane (PDMS) membranes were prepared for the VOC extraction studies. The influences of operating conditions (i.e., feed concentration and temperature) on the pervaporation performance of the membranes were investigated. In binary VOC/water mixtures, an increase in the feed concentration increased the VOC flux and decreased the separation factor. The VOC flux also increased significantly with temperature, mainly due to an augmented driving force for permeation. The impact of the coupling effects in multicomponent permeation was evaluated by comparing the pervaporation performance of VOCs in binary VOC/water and quaternary VOCs/water systems. Results show that the VOC permeation behavior was affected by the presence of other VOCs, depending on the permeant-permeant and membrane-permeant interactions. Based on pervaporation separation index, the PEBA membrane showed a better overall separation efficiency than the PDMS membrane for the extraction of perilla VOCs. Since pervaporation does not involve any chemical solvents and operates at moderate temperatures, it provides a green process for extracting valuable perilla VOCs.
