((2R,3S,4R,5R)-5-(3-Carbamoylpyridin-1-ium-1-yl)-3,4-dihydroxytetrahydrofuran-2-yl)methyl hydrogen phosphate (cas: 1094-61-7) belongs to amides. The amide group is called a peptide bond when it is part of the main chain of a protein, and an isopeptide bond when it occurs in a side chain, such as in the amino acids asparagine and glutamine. Amides are not in general accessible by the direct condensation of amines with carboxylic acids for two reasons: first, both components are readily deactivated by a transfer of a proton from the acid to the amine and second, the hydroxy unit on the carbonyl of the acid is a relatively poor leaving group. A growing body of evidence suggests that gut microbiota could participate in the progression of depression via the microbiota-gut-brain axis. However, the detailed microbial metabolic profile changes in the progression of depression is still not fully elucidated. In this study, a liquid chromatog. coupled to mass spectrometrybased untargeted serum high-throughput metabolomics method was first performed to screen for potential biomarkers in a depressive-like state in a chronic unpredictable mild stress (CUMS)-induced mouse model. Our results identified that the bile acid and energy metabolism pathways were significantly affected in CUMS progression. The detailed bile acid profiles were subsequently quantified in the serum, liver, and feces. The results showed that CUMS significantly promoted the deconjugation of conjugated bile acid and secondary bile acid biosynthesis. Furthermore, 16S rRNA gene sequencing revealed that the increased secondary bile acid levels in the feces pos. correlated with Ruminococcaceae_UCG-010, Ruminococcus, and Clostridia_UCG-014 abundance. Taken together, our study suggested that changes in family Ruminococcaceae abundance following chronic stress increased biosynthesis of deoxycholic acid (DCA), a unconjugated secondary bile acid in the intestine. Aberrant activation of secondary bile acid biosynthesis pathway thereby increased the hydrophobicity of the bile acid pool, which might, in turn, promoted metabolic disturbances and disease progression in CUMS mice. Nicotinamide mononucleotide attenuates isoproterenol-induced cardiac fibrosis by regulating oxidative stress and Smad3 acetylation was written by Wu, Keke;Li, Biao;Lin, Qiuzhen;Xu, Wanghan;Zuo, Wanyun;Li, Jiayi;Liu, Na;Tu, Tao;Zhang, Baojian;Xiao, Yichao;Liu, Qiming. Cardiac fibrosis is a pathol. hallmark of progressive heart diseases currently lacking effective treatment. NMN (NMN), a member of the vitamin B3 family, is a defined biosynthetic precursor of NAD (NAD+). Its beneficial effects on cardiac diseases are known, but its effects on cardiac fibrosis and the underlying mechanism remain unclear. We aimed to elucidate the protective effect of NMN against cardiac fibrosis and its underlying mechanisms of action. Cardiac fibrosis was induced by isoproterenol (ISO) in mice. NMN was administered by i.p. injection. In vitro, cardiac fibroblasts (CFs) were stimulated by transforming growth factor-beta (TGF-β) with or without NMN and sirtinol, a SIRT1 inhibitor. Levels of cardiac fibrosis, NAD+/SIRT1 alteration, oxidative stress, and Smad3 acetylation were evaluated by real-time polymerase chain reaction, western blots, immunohistochem. staining, immunoprecipitation, and assay kits. ISO treatment induced cardiac dysfunction, fibrosis, and hypertrophy in vivo, whereas NMN alleviated these changes. Addnl., NMN suppressed CFs activation stimulated by TGF-β in vitro. Mechanistically, NMN restored the NAD+/SIRT1 axis and inhibited the oxidative stress and Smad3 acetylation induced by ISO or TGF-β. However, the protective effects of NMN were partly antagonized by sirtinol in vitro. NMN could attenuate cardiac fibrosis in vivo and fibroblast activation in vitro by suppressing oxidative stress and Smad3 acetylation in a NAD+/SIRT1-dependent manner. In this study, the microwave-assisted processing of Sesamum indicum leaves was intensified to predict the total alkaloid content via Taguchi orthogonal design and neural network model. Under the optimum microwave condition, the maximized alkaloid content was estimated to be 15.40 (mg/g) % using irradiation time (A) of 60 s, microwave power (B) of 300 W, oven temperature (C) of 60°C, the mass ratio (D) of 12.5 g/mL, and solvent concentration (E) of 80%. The significant effects of each microwave extraction parameter declined in the order listed: C > B > A > D > E which indicated a 31 and 29% contribution of microwave power and oven temperature, resp. Moreover, higher coefficients of determination (R2 = 0.9999), and a decreased mean square error (MSE) of 2.69E-07 suggest good accuracy in predicting total bioactive alkaloid recovery, as proven by cross-validation trials. Under these microwave conditions, a total of 115 alkaloids were tentatively identified with lindelofine (m/z = 308.18) and supinin (m/z = 306.17) being the most abundant pyrrolizidine alkaloids. Although many researchers reported the total alkaloids recoveries from different parts of Sesame plant; the process intensification of microwave processing from leafy parts using Taguchi design and ANN approach have not been investigated. NAD (NAD +) acts as a cofactor for multiple biol. processes. While previous research has revealed that the NAD + declines associated with aging contributes to an impairment of immune cells, its role in mast cell function, especially in response to an anaphylactic condition, has remained unexplored. Author tested whether the restoration of cellular NAD + concentration by the supplementation of NAD + boosting mols. prevented mast cell degranulation and anaphylactic responses. Bone marrow derived mast cells (BMMCs) and human cord blood derived mast cells were treated with NAD + precursors NMN (NMN) and nicotinamide riboside (NR), and FceRI downstream signaling was assessed. Animal models of passive systemic anaphylaxis (PSA) and passive cutaneous anaphylaxis (PCA) were used to investigate the effects of NAD + precursors in the anaphylactic responses of mice. Treatment of murine BMMCs and human cord blood derived mast cells with NAD + precursors repressed intracellular signaling downstream of FceRI, as well as the release of inflammatory cytokines and lipid mediators. The i.p. administration of NMN or NR also markedly attenuated IgE-mediated anaphylactic responses in mouse models of PSA and PCA. These beneficial effects of NAD + precursors, however, were attenuated in mast cell-specific Sirt6 knockout mice, indicating a Sirt6 dependency for their action. NAD + precursors may serve as an effective therapeutic strategy that limits mast cell-mediated anaphylactic responses. It is known that the activities of nicotine adenine dinucleotide (NAD+)-dependent deacetylase decline in the aging mouse liver, and NMN (NMN)-mediated activation of deacetylase has been shown to increase healthspans. However, age-induced changes of the acetylomic landscape and effects of NMN treatment on protein acetylation have not been reported. Here, we performed immunoprecipitation coupled with label-free quant. LC-MS/MS (IPMS) to identify the acetylome and investigate the effects of aging and NMN on liver protein acetylation. In total, 7773 acetylated peptides assigned to 1997 proteins were commonly identified from young and aged livers treated with vehicle or NMN. The major biol. processes associated with proteins exhibiting increased acetylation from aged livers were oxidation-reduction and metabolic processes. Proteins with decreased acetylation from aged livers mostly participated in transport and translation processes. Furthermore, NMN treatment inhibited the aging-related increase of acetylation on proteins regulating fatty acid β oxidation, the tricarboxylic acid (TCA) cycle and valine degradation In particular, NAD (P) transhydrogenase (NNT) was markedly hyperacetylated at K70 in aged livers, and NMN treatment decreased acetylation intensity without altering protein levels. Mitochondria need to interact with the nucleus under homeostasis and stress to maintain cellular demands and nuclear transcriptional programs. Disrupted mitonuclear interaction is involved in many disease processes. However, the role of mitonuclear signaling regulators in endotoxin-induced acute lung injury (ALI) remains unknown. NAD (NAD+) is closely related to mitonuclear interaction with its central role in mitochondrial metabolism In the current study, C57BL/6J mice were administrated with lipopolysaccharide 15 mg/kg to induce endotoxin-induced ALI and investigated whether the NAD+ precursor NMN (NMN) could preserve mitonuclear interaction and alleviate ALI. After pretreatment with NMN for 7 days, NAD+ levels in the mitochondrial, nucleus, and total intracellular were significantly increased in endotoxemia mice. Moreover, supplementation of NMN alleviated lung pathol. injury, reduced ROS levels, increased MnSOD activities, mitigated mitochondrial dysfunction, ameliorated the defects in the nucleus morphol., and these cytoprotective effects were accompanied by preserving mitonuclear interaction (including mitonuclear protein imbalance and the mitochondrial unfolded protein response, UPRmt). Furthermore, NAD+-mediated mitonuclear protein imbalance and UPRmt are probably regulated by deacetylase Sirtuin1 (SIRT1). The presence of the amide group –C(=O)N– is generally easily established, at least in small molecules. It can be distinguished from nitro and cyano groups in IR spectra. Amides exhibit a moderately intense νCO band near 1650 cm−1. By 1H NMR spectroscopy, CONHR signals occur at low fields. In X-ray crystallography, the C(=O)N center together with the three immediately adjacent atoms characteristically define a plane.Recommanded Product: 1094-61-7

Anderson, Jan M. et al. published their research in Biochimica et Biophysica Acta, Biophysics Including Photosynthesis in 1966 | CAS: 730-25-6

N-(3,4-Dichlorophenyl)octanamide (cas: 730-25-6) belongs to amides. Compared to amines, amides are very weak bases and do not have clearly defined acid–base properties in water. On the other hand, amides are much stronger bases than esters, aldehydes, and ketones. Amides are stable compounds. The lower-melting members (such as acetamide) can be readily purified by fractional distillation. Most amides are solids which have low solubilities in water.Reference of 730-25-6

Fractionation of the photochemical systems of photosynthesis. I. Chlorophyll contents and photochemical activities of particles isolated from spinach chloroplasts was written by Anderson, Jan M.;Boardman, N. K.. And the article was included in Biochimica et Biophysica Acta, Biophysics Including Photosynthesis in 1966.Reference of 730-25-6 The following contents are mentioned in the article:

Spinach chloroplasts were fragmented by incubation with digitonin and the resulting chlorophyll-containing particles separated by differential centrifugation. Particles pelleted by forces of 1000 g and 10,000 g had a lower ratio of chlorophyll a/chlorophyll b (chl a/chl b) than the chloroplasts, whereas the smaller particles which either sedimented at 50,000-144,000 g or remained in the 144,000-g supernatant had higher ratios than the chloroplasts. Chl a/chl b ratios were usually determined spectrophotometrically in 80% acetone, but essentially the same ratios were obtained after transfer of the pigments to ether or after separation of the chlorophylls by thin-layer chromatography. The small particles were inactive in the Hill reaction, either with ferricyanide, trichloroindophenol, or NADP as oxidant, but they photoreduced NADP if provided with the electron-donor couple, Na ascorbate, and dichloroindophenol, and both ferredoxin and NADP reductase. The 1000-g and 10,000-g fractions showed Hill activity, but the rate of reduction of NADP in the presence of ferredoxin and NADP reductase was lower than the rates of reduction of ferricyanide and trichloroindophenol. The particles with the high chl a/chl b ratios appear to be representative of System I (the “long wavelength” system) of photosynthesis, whereas the 1000-g and 10,000-g fractions appear to be enriched in particles representative of System 2. Chloroplasts were also fragmented by Triton X-100 and Nonidet P-40; in contrast to the digiton treatment, the particles so obtained were photochem. inactive. This study involved multiple reactions and reactants, such as N-(3,4-Dichlorophenyl)octanamide (cas: 730-25-6Reference of 730-25-6).

Althaus, Julia B. et al. published their research in Molecules in 2017 | CAS: 18836-52-7

(2E,4E)-N-Isobutyldeca-2,4-dienamide (cas: 18836-52-7) belongs to amides. Because of the greater electronegativity of oxygen, the carbonyl (C=O) is a stronger dipole than the N–C dipole. The presence of a C=O dipole and, to a lesser extent a N–C dipole, allows amides to act as H-bond acceptors. Amides are stable compounds. Alkamides from Anacyclus pyrethrum L. and their in vitro antiprotozoal activity was written by Althaus, Julia B.;Malyszek, Claudine;Kaiser, Marcel;Brun, Reto;Schmidt, Thomas J.. And the article was included in Molecules in 2017.Recommanded Product: 18836-52-7 The following contents are mentioned in the article:

In our ongoing study to evaluate the antiprotozoal activity of alkamides from Asteraceae, a dichloromethane extract from the roots of Anacyclus pyrethrum L. showed a moderate in vitro activity against the NF54 strain of Plasmodium falciparum and against Leishmania donovani (amastigotes, MHOM/ET/67/L82 strain). Seven pure alkamides and a mixture of two further alkamides were isolated by column chromatog. followed by preparative high performance liquid chromatog. The alkamides were identified by mass- and NMR-spectroscopic methods as tetradeca-2E,4E-dien-8,10-diynoic acid isobutylamide (anacycline, 1), deca-2E,4E-dienoic acid isobutylamide (pellitorine, 2), deca-2E,4E,9-trienoic acid isobutylamide (3), deca-2E,4E-dienoic acid 2-phenylethylamide (4), undeca-2E,4E-dien-8,10-diynoic acid isopentylamide (5), tetradeca-2E,4E,12Z-trien-8,10-diynoic acid isobutylamide (6), and dodeca-2E,4E-dien acid 4-hydroxy-2-phenylethylamide (7). Two compounds-undeca-2E,4E-dien-8,10-diynoic acid 2-phenylethylamide (8) and deca-2E,4E-dienoic acid 4-hydroxy-2-phenylethylamide (9) -were isolated as an inseparable mixture (1:4). Compounds 3, 4, and 5 were isolated from Anacyclus pyrethrum L. for the first time. While compounds 4 and 5 were previously known from the genus Achillea, compound 3 is a new natural product, to the best of our knowledge. All isolated alkamides were tested in vitro for antiprotozoal activity against Plasmodium falciparum, Trypanosoma brucei rhodesiense, Trypanosoma cruzi, and Leishmania donovani and for cytotoxicity against L6 rat skeletal myoblasts.
