N-aromatic acyl-amino acid conjugates possess a colon-targeted property, implying that such conjugates are stable and are not absorbable until reaching the large intestine in which they are microbially converted (hydrolyzed) to the parent drugs that are therapeutically active. To investigate the structural effect of N-aromatic acyl-amino acid conjugates on the large intestinal deconjugation, the hydrolysis of various N-aromatic acyl-amino acid conjugates was examined in the cecal contents. On incubation of conjugates with glycine, D or/andL forms of alanine or phenylalanine in the cecal contents, the conjugates with D amino acids were not hydrolyzed. The other conjugates are susceptible to the hydrolysis, the rates of which decreased as the size of the substituent on the 2-position of the amino acids increased. The conjugates with alkyl analogs (2-4 carbons) of glycine and taurine were resistant to the hydrolysis, while taurine- and glycine-conjugates were hydrolyzed effectively. The hydrolysis of N-aromatic acyl-glycine conjugates was enhanced by para-substitution of electron withdrawing groups on the aromatic acyl moiety and vice versa for electron-donating groups. While a Me, methoxy or chloro group on the ortho-position retarded the hydrolysis, a hydroxyl group on the position accelerated it. A dynamic pH junction was used in capillary electrophoresis (CE-DAD) to online preconc., sep., and determine trace amounts of sulfonamide antibiotics (SAs) in milk and yoghurt samples in this study. A sample matrix with 0.15% acetic acid and 10% methanol (MeOH) at a pH of 4.0, and a background electrolyte (BGE) that contained 35 mM sodium citrate with 10% MeOH at a pH of 8.5, and an acidic barrage of 0.4% acetic acid with 10% MeOH at a pH of 2.5 were utilized to achieve a stacking effect for SAs through a dynamic pH junction. Under optimized conditions, the proposed preconcentration method showed good linearity (30-500 ng/mL, r2 ≥ 0.9940), low limits of detection (LODs) of 4.1-6.3 ng/mL, and acceptable analytes recovery (81.2-106.9%) with relative standard deviations (RSDs) within 5.3-13.7 (n = 9). The limits of quantification (LOQs) were below the maximum residue limit approved by the European Union (EU) in this type of matrixes. Sensitivity enhancement factors of up to 129 were reached with the optimized dynamic pH junction using CE with a diode array detector (DAD). The method was used to determine SAs in fresh milk, low-fat milk, full-cream milk, and yoghurt samples. In this study, a powerful and reliable strategy for high-throughput screening of sulfonamide metabolites in goat meat was proposed based on an aqueous two-phase separation procedure (ATPS) combined with ultrahigh-performance liquid chromatog. quadrupole-Orbitrap high-resolution mass spectrometry (UHPLC-Q-Orbitrap). Noncovalent interactions including van der Waals force, hydrogen bonding, and hydrophobic effect were determined to be staple interactions between the sulfonamide metabolites and sheep serum albumin by fluorescence spectroscopy and mol. docking technol., and an 80% acetonitrile-water solution/(NH4)2SO4 was used as ATPS in order to release combined sulfonamide metabolites and minimize the influence of sheep serum albumin. Sulfonamide metabolites in the matrix were screened based on a mechanism of mass natural loss and core structure followed by identification combined with the pharmacokinetic. The developed strategy was validated according to EU standard 2002/657/EC with CCα ranging from 0.07 to 0.98μg kg-1, accuracy recovery with 84-107%, and RSDs lower than 8.9%. Eighty seven goat meat samples were used for determination of 26 sulfonamides and 8 potential metabolites. On the basis of the established innovative process, this study has successfully implemented the comprehensive detection of sulfonamide metabolites, including N4-acetylated substitution, N4-hydroxylation, 4-nitroso, azo dimers, oxidized nitro, N4 monoglucose conjugation, β-D-glucuronide, and N-4-aminobenzenesulfonyl metabolites, which were shown to undergo oxidation, hydrogenation, sulfation, glucuronidation, glucosylation, and O-aminomethylation. The purpose of this study was to develop a modified QuEChERS method based on melamine sponge for rapid determination of multi-class veterinary drugs in milks by UPLC-MS/MS. Through simple infiltration and extrusion, fast and convenient matrix purification could be achieved within several seconds, and there was no need of extra phase separation operations. Good linearity with correlation coefficient (R2) ≥0.999 was obtained for all drugs in the range of 2∼500μg·kg-1. The obtained matrix effects were within ±20% for all monitored drugs. The recoveries of all monitored drugs ranged from 60.7% to 116.0% at three spiked levels (50, 100, and 200μg·kg-1), with relative standard deviations less than 7.4%. Comparatively low LODs and LOQs were obtained in the ranges of 0.1∼3.8μg·kg-1 and 0.2∼6.3μg·kg-1, resp. Compared with conventional purification adsorbents, melamine sponge yielded an equal or higher purification performance with matrix removal rate as high as 52.5% and acceptable recoveries in range of 60%-120% for all monitored drugs. The satisfactory results have demonstrated the good potential of melamine sponge in matrix purification for rapid determination of multiclass residues in food safety. Suspect screening was conducted by comparing the exptl. measured exact mass of precursor and fragment ions with the custom accurate-mass database and by matching the acquired mass spectra against the mass spectral library. The anal. performance of the developed methodol. was assessed in terms of sensitivity, linearity, repeatability, and recovery. The limits of detection (LODs) and quantitation (LOQs) of the 52 analytes were in the range of 0.5-20μg/L and 2-50μg/L, resp. The matrix effect ratios ranged from -23.5% to 25.9%.
