The large-scale synthetic route to the macrocycle employed a unique intramolecular etherification reaction as the key step in the macrocycle synthesis, avoiding the scalability limitations of the ring-closing metathesis (RCM) reaction of the enabling route. The large-scale synthetic route to the macrocycle was successfully used to produce the amount of glecaprevir required to support the late-stage clinical development.

To explore new skeleton with nematicidal activity, a series of novel 1,2,3-benzotriazin-4-one derivatives containing piperazine as linker were synthesized and varied fragments were also introduced to increase structure diversity of the new skeleton. Their inhibitory activities in vivo were evaluated against Meloidogyne incognita. The newly prepared compounds A6, A8, A21, A28 and A38 exhibited more than 50% inhibition at the concentration of 20 mg/L. Especially compound A6 displayed 71.4% inhibition against Meloidogyne incognita at the concentration of 20 mg/L. The nematicidal activities varied significantly depending on the types and positions of the substituents, which provided guidance for further structure modification. Insect-microbial symbioses have vast biochemical diversity, which is beneficial to produce bioactive secondary metabolites. In this study, chemical examination of a Streptomyces sp. associated with a mud dauber wasp led to the isolation of fourteen compounds. Their structures were determined by spectroscopic methods and comparison with literature data. Among the isolates, compounds 1,2,3-benzotriazin-4(1H)-one and 4-(2-aminoethyl)phenyl acetate were first reported from this species. Bioactivities of the isolated compounds were assayed for the first time against hexokinase II. 4-(2-Aminoethyl)phenyl acetate, germicidin B, phenylacetic acid, isogermicidin A and germicidin C displayed significant inhibitory activity against hexokinase II, with the IC50 values of 5.11, 7.11, 7.15, 8.45 and 8.78 mu M, respectively.

An efficient Ru-catalyzed asymmetric hydrogenation of dibenzo[c,e]azepines is reported. A series of seven-membered cyclic amines were obtained with moderate to excellent enantioselectivity. The catalyst counteranion played an important role in achieving high-level chiral induction. Moreover, a one-pot synthesis of chiral 6,7-dihydro-5H-dibenz[c,e]azepines via two-step reductive amination was also developed.

Without using chiral sources, the Strecker reaction of achiral hydrogen cyanide, p-tolualdehyde and benzhydrylamine gave enantioenriched L- or D-N-benzhydryl-alpha-( p-tolyl) glycine nitriles with up to > 99% ee in a mixed solvent of water and methanol. Therefore, total spontaneous resolution of alpha-aminonitriles could occur through a prebiotic mechanism of alpha-amino acid synthesis. Moreover, it was demonstrated that the repetition of partial dissolution and crystallization of a suspended conglomerate of aminonitrile under solution-phase racemization could generate the enantiomeric imbalance to afford, in combination with the amplification of chirality, an enantioenriched product in every case. Among the 73 experiments that were carried out, D-and L-enriched isomers occurred 36 and 37 times, respectively. This stochastic behavior, under achiral or racemic starting conditions, meets the requirements of the spontaneous absolute asymmetric Strecker synthesis. The implications of the present results for the origin of chirality of alpha-amino acids are discussed.

In this paper, we report on a new dielectric, a C-60-containing block polyimide (PI-b-C-60). This was realized by introducing C-60 as pendent groups into a polymer backbone. When this dielectric was used in a non-contact mode triboelectric nanogenerator (TENG), it achieved high output power and reliable operation. Compared with perfluoroalkoxy alkane film-based TENGs, the TENG based on PI-b-C-60 generated 4.3 times higher output power and a superior charge density of over 300 mu C m(-2) with a 3 times slower charge decay rate. These results are most likely due to the excellent charge-retention characteristics induced by the most negative electrostatic potential of C-60 within the backbone, and these characteristics were confirmed by surface potential measurements. Furthermore, in the course of our work, two non-contact mode applications, a keyless electronic door lock system and a speed sensor with a tone wheel for a car, were developed. Without an ion injection process being needed, very sensitive and reliable operations of the speed sensor were successfully demonstrated, even under very humid conditions (similar to 99% RH).

A simple and efficient protocol for the formylation of amines with formic acid, catalyzed by a polyoxometalate-based chromium catalyst, is described. Notably, this method shows excellent activity and chemo-selectivity for the formylation of primary amines; diamines have also been successfully employed. Importantly, the chromium catalyst is potentially non-toxic, environmentally benign and safer than the widely used high valence chromium catalysts such as CrO3 and K2Cr2O7.
