The mechanical stability of conventional single-channel capillary fibres can be improved in a multi-channel geometry, which has previously found application in ultrafiltration. In this work, multi-channel polyethersulfone (PES) capillary membranes comprising seven feed channels were successfully fabricated in an enhanced steam-dry-wet spinning process and coated on the inner surface with a thin polyamide (PA) layer via interfacial polymerization (IP). The coating procedure consisted of impregnating the support multi-channel capillary membranes (MCM) with an aqueous piperazine solution, flushing with nitrogen gas to remove excess droplets, and pumping an organic trimesoylchloride solution through the channels. Insights into the interfacial polymerization process were gained through the investigation of various parameters, including monomer ratio, contact time, and drying time. Membranes were characterised via scanning electron microscopy (SEM), atomic force microscopy (AFM), and filtration experiments. The optimisation of both the PES support membrane and IP process parameters allowed for the fabrication of composite MCM with an MgSO4 rejection of 91.4% and a solute flux of 68.8 L m(-2) h(-1) at an applied pressure of 3 bar. The fabricated composite MCM demonstrates that a favourable multi-channel arrangement can be upgraded with a PA layer for application in low-pressure nanofiltration.

The ratio of the bonding water state (i.e., intermediate and nonfreezing water molecules) is almost the same between two HAp films, and the E-HAp film with an elliptical shape and smaller particle size exhibited the smallest ratio of nonfreezing water, which can suppress the denaturation of the adsorbed protein. Subsequently, FT-IR spectral deconvolution results of the amide I band of the adsorbed Fgn on the E-HAp film indicated the higher proportion of a-helix and beta-sheet structures as compared with those on the N-HAp and Au films, suggesting that the smaller proportion of nonfreezing waters would play a significant role in the stereoscopic Fgn conformation. In the culture of fibroblasts, FT-IR spectra of the adhered cells on the E-HAp, N-HAp, and Au films exhibited different absorbance intensities of the amide A, I, II, and III bands, suggesting a different amount of collagen-producing states by the cells, which were also supported by immunostaining results of the collagen type I. Therefore, the different hydration structures on the films clearly influenced the conformation of the adsorbed protein, and the preferential conformation was found at the interfaces between the fibroblasts and the underground E-HAp films.

Seed aging affects the agricultural production and quality of food. Therefore, it is of great significance to distinguish the aging seed quickly for food security. In this work, Fourier transform infrared (FTIR) spectroscopy combined with curve-fitting analysis and hierarchical cluster analysis (HCA) was used to identify the legume seeds of different aging time. The results showed that the infrared spectroscopy of legume seeds were mainly composed of the absorption peaks of proteins and carbohydrates. The overall characteristics of the original spectra of seeds in different aging time were similar, but the absorption ratios of several peaks decreased with the increasing aging time. Amide I band (1700-1600 cm(-1)) and carbohydrate absorption band (1180-980 cm(-1)) in the original spectra were carried out for curve fitting. The results showed that the sub-bands position and area ratios of different aging seeds were obviously different. There were significant differences in beta-folding, disordered structure, alpha-helix and beta-corner components in the secondary structure of proteins and C-O, C-C and C-O-H components in polysaccharides during aging. Hierarchical cluster analysis was performed using second derivative spectra in the range of 1800-800 cm(-1), and the accuracy of clustering reached 100%. The results showed that FTIR combined with curve. fitting analysis and HCA could identify the natural aging legume seeds quickly and effectively.

Mechanisms of protein-carbohydrate recognition attract a lot of interest due to their roles in various cellular processes and metabolism disorders. We have performed a large-scale analysis of protein structures solved in complex with glucose, galactose and their substituted analogues. We found that, on average, sugar molecules establish five hydrogen bonds (HBs) in the binding site, including one to three HBs with bridging water molecules. The free energy contribution of bridging and direct HBs was estimated using the free energy perturbation (FEP+) methodology for mono- and disaccharides that bind to l-ABP, ttGBP, TrmB, hGalectin-1 and hGalectin-3. We show that removing hydroxy groups that are engaged in direct HBs with the charged groups of Asp, Arg and Glu residues, protein backbone amide or buried water dramatically decreases binding affinity. In contrast, all solvent-exposed hydroxy groups and hydroxy groups engaged in HBs with the solvent-exposed bridging water molecules contribute weakly to binding affinity and so can be replaced to optimize ligand potency. Finally, we rationalize an effect of binding site water replacement on the binding affinity to l-ABP.
