The optical density changes (Delta OD) were observed in the range of 0.18-0.68 at specific absorption maxima, with the calculated coloration efficiencies of 42-123 cm(2)/C. Single-layer electrochromic devices using the electrodeposited polymer films as active layers were fabricated for the preliminary investigation of their electrochromic applications.

Novel Homo-Bivalent and Polyvalent Compounds Based on Ligustrazine and Heterocyclic Ring as Anticancer Agents

Bivalent and polyvalent inhibitors can be used as antitumor agents. In this experiment, eight ligustrazine dimers and seven ligustrazine tetramers linked by alkane diamine with different lengths of carbon chain lengths were synthesized. After screening their antiproliferation activities against five cancer cell lines, most ligustrazine derivatives showed better cytotoxicity than the ligustrazine monomer. In particular, ligustrazine dimer 8e linked with decane-1,10-diamine exhibited the highest cytotoxicity in FaDu cells with an IC50 (50% inhibiting concentration) value of 1.36 nM. Further mechanism studies suggested that 8e could induce apoptosis of FaDu cells through the depolarization of mitochondrial membrane potential and S-phase cell cycle arrest. Inspired by these results, twenty-seven additional small molecule heterocyclic dimers linked with decane-1,10-diamine and nine cinnamic acid dimers bearing ether chain were synthesized and screened. Most monocyclic and bicyclic aromatic systems showed highly selective anti-proliferation activity to FaDu cells and low toxicity to normal MCF 10A cells. The structure-activity relationship revealed that the two terminal amide bonds and the alkyl linker with a chain length of 8-12 carbon were two important factors to maintain its antitumor activity. In addition, the ADMET calculation predicted that most of the potent compounds had good oral bioavailability.

The investigation of detection and sensing mechanism of spicy substance based on human TRPV1 channel protein-cell membrane biosensor

The transient receptor potential vanilloid 1 (TRPV1) is a key target for the spicy taste sensor and analgesic drug development. However, the human TRPV1-associated signaling remains to be obscure. In this study, we over expressed human TRPV1 (hTRPV1) in HEK293T cells and explored its signaling activated by spicy substances. A cell membrane biosensor was constructed by using the cells highly expressed hTRPV1 through a layer-by-layer assembly. Our results showed that the activation constants by capsaicin, allicin and sanshool, the active components of chili pepper, garlic and mountain pepper, were Ka, capsaicin = 3.5206 x 10(-16) mol/L, Ka, allicin = 5.0227 x 10(-15) mol/L, Ka, sanshool = 1.7832 x 10(-15) mol/L. Obviously, the order of the sensitivity mediated by hTRPV1 was capsaicin > sanshool > allicin. The affinity values of the three spicy substances with hTRPV1 analyzed by molecular docking simulation also displayed the same law. Most importantly, some amide bonds and their similar groups and even benzene rings of spicy compounds were fund to be critical in the spicy sensing process. In addition, Glu570 in the active pocket of hTRPV1 plays an important role in identifying spicy substances. The elucidation of the detailed mechanism mediated by hTRPV1 in spicy sensing will lay a theoretical foundation to design rational strategies for screening of potential analgesics.

Catalytic Enantioselective Synthesis of 3,4-Polyfused Oxindoles with Quaternary All-Carbon Stereocenters: A Rh-Catalyzed C-C Activation Approach

The first Rh-catalyzed enantioselective synthesis of a 3,4-polyfused oxindole ring system enabled by carboacylation of acrylic amides based on C-C activation is reported. Estimation of Glucose Content in Raw Potatoes with a Biosensor as an Indicator of Acrylamide Level in Processed Potatoes

Acrylamide is an amide-type organic compound formed by the reaction of asparagine with reducing sugars (Maillard reaction) and is classified as a probable human carcinogen (Group 2A) by the International Agency for Research on Cancer. Potato chips are one of the foods that contribute most to dietary acrylamide intake, and despite a significant reduction in recent years thanks to changes in frying methods, many commercial samples show concentrations above the levels recommended by the European Commission Regulation 2017/2158. In this work, a high correlation (P<0.01) was established (r = 0.9592) between the content of reducing sugars and glucose measured by the BIOWINE 700 GLU (R) biosensor from Biolan Microbiosensors S.L. A high correlation was also obtained between the glucose content of the raw potato and the acrylamide level of potato chips fried at 150 degrees C (r = 0.8985, P<0.01) and 176 degrees C (r = 0.9949, P<0.01). The lengths of Si-F-ax. comprise 1.686, 1.661 and 1.632 angstrom respectively. QTAIM analysis was used to estimate the energy of co-ordination bond C=O -> Si in these amides. Fatty acid amide hydrolase (FAAH) degrades 2 major classes of bioactive fatty acid amides, the N-acylethanolamines (NAEs) and N-acyl taurines (NATs), in central and peripheral tissues. A functional polymorphism in the human FAAH gene is linked to obesity and mice lacking FAAH show altered metabolic states, but whether these phenotypes are caused by elevations in NAEs or NATs is unknown. To overcome the problem of concurrent elevation of NAEs and NATs caused by genetic or pharmacological disruption of FAAH in vivo, we developed an engineered mouse model harboring a single-amino acid substitution in FAAH (S268D) that selectively disrupts NAT, but not NAE, hydrolytic activity. The FAAH-S268D mice accordingly show substantial elevations in NATs without alterations in NAE content, a unique metabolic profile that correlates with heightened insulin sensitivity and GLP-1 secretion. We also show that N-oleoyl taurine (C18:1 NAT), the most abundant NAT in human plasma, decreases food intake, improves glucose tolerance, and stimulates GPR119-dependent GLP-1 and glucagon secretion in mice. Together, these data suggest that NATs act as a class of lipid messengers that improve postprandial glucose regulation and may have potential as investigational metabolites to modify metabolic disease.

Metathesis in water conducted by tailor-made encapsulated Grubbs’ catalyst

Metathesis in water represents a current challenge in green chemistry, since hydrophobic catalysts are non-soluble in this medium. Although this issue has been addressed by modification of the hydrophobic ligand structure, alternative methods for conducting metathesis in water are of interest, such as catalyst encapsulation. In this contribution we report successful encapsulation of the Grubbs’ second-generation catalyst in alginate hydrogels, representing a renewable resource, to perform ring-closing metathesis (RCM) in water. We initially investigated the influence of different solvents on the reaction rate and confirmed that water is a preferred solvent. A comparison of non-encapsulated and encapsulated catalyst in calcium alginate revealed that the reaction rate for non-encapsulated catalyst was notably higher, which can be explained by on water conditions in this case. Inside the beads the encapsulated catalyst remained heterogeneous, thus allowing to switch the catalysis mode between in/on water through encapsulation. To overcome diffusion limitation and enhance reaction rate, we prepared a tailor-made bead material by introducing hydrophobic octyl-grafted alginate amide. Using such a hydrogel, diffusion limitation was positively influenced by hydrophobisation of the matrix, resulting in up to quadrupled reaction rates compared to calcium alginate as a standard encapsulation material. In terms of recycling, this encapsulated catalyst revealed promising performance, retaining 80% of its activity after ten runs. This is the first reported application of hydrophobised alginate derivatives in catalysed organic synthesis, achieving excellent encapsulation efficiency, no measurable leaching and yields of up to 87%.
