Reinforcing effect of aramid fibers on fatigue behavior of SBR/aramid fiber composites

This paper investigates the correlation between fatigue performance and static tensile behavior of fiber reinforced rubber materials and presents change of fiber reinforced effect during fatigue process. A rough aramid fiber (AF) surface, which is helpful for combining AF and rubber matrix, was achieved by etching AF in NaOH/CH3CH2OH solution via the hydrolysis of amide bonds on the fiber. Then, the treated fiber was coated with an epoxy-functionalized silane (gamma-propyltrimethoxysilane, KH560) and was used to prepare AF/carbon black/styrene-butadiene rubber (AF/CB/SBR) composites. The results showed that the interfacial adhesion between the complex-modified AF and the rubber matrix was further enhanced and the mechanical properties of the composite were further improved compared with those of the composite which was prepared by unhydrolyzed AF coated with KH560. The hysteresis reinforcing effect of the fiber was found after 30,000 cycles of strain-controlled fatigue, and the reinforcing effect of AF was reduced with increasing average fatigue strain. Moreover, the relative debonding energy (RDE) was calculated to evaluate the reinforcing effect of the fiber, and the RDE value sharply decreased after 30,000 cycles of strain-controlled fatigue. When the AF was treated with the NaOH/CH3CH2OH solution and then coated with KH560, the rubber composite had a high RDE value, and the fatigue life of the composite under the stress-control condition was increased approximately 4 times. The results from the characterization of the fracture appearance after 30,000 cycles of fatigue showed that the composite with the complex-modified AF could maintain better interface bonding during the fatigue process.

Synthesis of thermo- and photo-responsive polysiloxanes with tunable phase separation via aza-Michael addition

Two kinds of thermo- and photo-dual-responsive polysiloxanes (DRPSs) with functional pendent groups, N-isopropyl amides and azobenzene (Azo) or salicylideneaniline (SA), were synthesized through a facile, effective, and catalyst-free aza-Michael addition of poly(aminopropylmethyl-siloxane) with N-isopropyl acrylamide and N-azobenzene acrylamide or N-salicylaldehyde acrylamide. The chemical structrures of DRPSs were systematically characterized using FT-IR, H NMR and UV-Vis spectroscopy. The as-prepared DRPSs with lower Azo or SA contents exhibited lower critical solution temperature (LCST)-type phase transition in water, which is reversible and can be controlled by temperature and UV light. The effects of Azo and SA contents on the responsive properties of DRPSs are examined in detail. The LCST decreased with the increasing Azo or SA content. Once the content of Azo or SA reached up to 5.7% or 8.2%, respectively, DRPSs could not be dissolved in water even in an ice bath. Higher values of the LCST were measured after irradiation of the polymer solutions due to the higher polarity of cis-Azo and keto-SA conformation, induced by irradiation. The differences in cloud points between the irradiated and the non-irradiated DRPS aqueous solutions increased up to 3.4 degrees C and 9.8 degrees C when combined with 3.8% Azo and 5.8% SA units, respectively.

Hydrogen bonds (H-bonds) within the secondary coordination sphere are often invoked as essential noncovalent interactions that lead to productive chemistry in metalloproteins. Incorporating these types of effects within synthetic systems has proven a challenge in molecular design that often requires the use of rigid organic scaffolds to support H-bond donors or acceptors. We describe the preparation and characterization of a new hybrid tripodal ligand ([H(2)pout](3-)) that contains two monodeprotonated urea groups and one phosphinic amide. The urea groups serve as H-bond donors, while the phosphinic amide group serves as a single H-bond acceptor. The [H(2)pout](3-) ligand was utilized to stabilize a series of Mn-hydroxido complexes in which the oxidation state of the metal center ranges from 2+ to 4+. The molecular structure of the Mn-III-OH complex demonstrates that three intramolecular H-bonds involving the hydroxido ligand are formed. Additional evidence for the formation of intramolecular H-bonds was provided by vibrational spectroscopy in which the energy of the O-H vibration supports its assignment as an H-bond donor. The stepwise oxidation of [Mn(II)H(2)pout(OH)](2-) to its higher oxidized analogs was further substantiated by electrochemical measurements and results from electronic absorbance and electron paramagnetic resonance spectroscopies. Our findings illustrate the utility of controlling both the primary and secondary coordination spheres to achieve structurally similar Mn-OH complexes with varying oxidation states.

Interfacial Effect of Hydration Structures of Hydroxyapatite Nanoparticle Films on Protein Adsorption and Cell Adhesion States

The synthesized elliptical hydroxyapatite (E-HAp) and needle-like HAp (N-HAp) nanoparticles (NPs) were electrophoretically deposited on a gold (Au) substrate. A comparative study of the hydration layers on E-HAp, N-HAp, and Au films was achieved to investigate the interfacial effect of the hydration layers on the conformation of the adsorbed fibrinogen (Fgn) and fibroblast adhesion properties. As a result, the ratios of three types of hydration layer states (free water, intermediate water, nonfreezing water) analyzed by a Fourier transform infrared (FT-IR) spectral deconvolution of the O-H stretching absorption band were investigated. The ratio of the bonding water state (i.e., intermediate and nonfreezing water molecules) is almost the same between two HAp films, and the E-HAp film with an elliptical shape and smaller particle size exhibited the smallest ratio of nonfreezing water, which can suppress the denaturation of the adsorbed protein. Subsequently, FT-IR spectral deconvolution results of the amide I band of the adsorbed Fgn on the E-HAp film indicated the higher proportion of a-helix and beta-sheet structures as compared with those on the N-HAp and Au films, suggesting that the smaller proportion of nonfreezing waters would play a significant role in the stereoscopic Fgn conformation. In the culture of fibroblasts, FT-IR spectra of the adhered cells on the E-HAp, N-HAp, and Au films exhibited different absorbance intensities of the amide A, I, II, and III bands, suggesting a different amount of collagen-producing states by the cells, which were also supported by immunostaining results of the collagen type I. Therefore, the different hydration structures on the films clearly influenced the conformation of the adsorbed protein, and the preferential conformation was found at the interfaces between the fibroblasts and the underground E-HAp films.

Amino acids are attached to the tRNA 3′-end as a prerequisite for entering the ribosome for protein synthesis. Amino acid attachment also gives tRNA access to nonribosomal cellular activities. However, the normal attachment is via an ester linkage between the carboxylic group of the amino acid and the 3′-hydroxyl of the terminal A76 ribose in tRNA. The instability of this ester linkage has severely hampered studies of aminoacyl-tRNAs. Although the use of 3′-amino-3′-deoxy A76 in a 3’amino-tailed tRNA provides stable aminoacyl attachment via an amide linkage, there are multiple tailing protocols and the efficiency of each relative to the others is unknown. Here we compare five different tailing protocols in parallel, all dependent on the CCA-adding enzyme [CTP(ATP): tRNA nucleotidyl transferase; abbreviated as the CCA enzyme] to exchange the natural ribose with the modified one. We show that the most efficient protocol is achieved by the CCA-catalyzed pyrophosphorolysis removal of the natural A76 in equilibrium with the addition of the appropriate ATP analog to synthesize the modified 3′-end. This protocol for 3’-amino-tailing affords quantitative and stable attachment of a broad range of amino acids to tRNA, indicating its general utility for studies of aminoacyl-tRNAs in both canonical and noncanonical activities.

Lepadins I-K, 3-O-(3 ‘-Methylthio)acryloyloxy-decahydroquinoline Esters from a Bahamian Ascidian Didemnum sp Assignment of Absolute Stereostructures

Three decahydroisoquinoline alkaloids, lepadins I-K, were isolated from a specimen of Didemnum sp. collected in the Bahamas. The structures of the new compounds were assigned by an integrated analysis of MS, IR, and H-1, C-13, and 2D NMR spectra. Like previously reported lepadins, the structures of the new compounds contain a decahydroquinoline heterocyclic core in lepadin I, and a new variation, an octahydroquinoline in lepadin J, but differ from earlier reported compounds by acylation of the 3-hydroxyl group by a rare 3’-methylthioacrylate. The absolute configuration of lepadin I was solved by interpretation of NOE measurements, and exciton coupled circular dichroism (ECCD) of the corresponding N-p-bromobenzoyl derivative. The latter constitutes a general method for determination of absolute configuration of the entire lepadin family. The configuration of the remote side-chain secondary carbinol was solved by the modified Mosher’s esters method. Lepadin I inhibited butyrylcholineesterase (BuChE, IC50 3.1 mu M), but only weakly inhibited acetylcholineesterase (AChE) (10% at 100 mu M).
