2-Amino-benzo[d]thiazole was identified as a new scaffold for the development of improved pteridine reductase-1 (PTR1) inhibitors and anti-trypanosomatidic agents. Molecular docking and crystallography guided the design and synthesis of 42 new benzothiazoles. The compounds were assessed for Trypanosoma brucei and Leishmania major PTR1 inhibition and in vitro activity against T. brucei and amastigote Leishmania infantum. We identified several 2-amino-benzo[d]thiazoles with improved enzymatic activity (TbPTR1 IC50 = 0.35 mu M; LmPTR1 IC50 = 1.9 mu M) and low mu M antiparasitic activity against T. brucei. The ten most active compounds against TbPTR1 were able to potentiate the antiparasitic activity of methotrexate when evaluated in combination against T. brucei, with a potentiating index between 1.2 and 2.7. The compound library was profiled for early ADME toxicity, and 2-amino-N-benzylbenzo[d]thiazole-6-carboxamide (4c) was finally identified as a novel potent, safe, and selective anti-trypanocydal agent (EC50 = 7.0 mu M). Formulation of 4c with hydroxypropyl-beta-cyclodextrin yielded good oral bioavailability, encouraging progression to in vivo studies.

The isomerization of epoxides to aldehydes using the readily available Crabtree’s reagent is described. The aldehydes were transformed into synthetically useful amines by a one-pot reductive amination using pyrrolidine as imine-formation catalyst. The reactions worked with low catalyst loadings in very mild conditions. The procedure is operationally simple and tolerates a wide range of functional groups. A DFT study of its mechanism is presented showing that the isomerization takes place via an iridium hydride mechanism with a low energy barrier, in agreement with the mild reaction conditions.

The aldol reaction which is the most important one among the C-C bond forming reactions, is widely used by synthetic organic chemists to obtain beta-hydroxycarbonyl compounds which are important starting components for biologically active compounds in optically pure form. In this research, five Pro-Phe derivatives were synthesized by simple amidation reactions and characterized by their spectral data. Their catalytic activities in asymmetric aldol reaction were investigated. The catalytic activity studies were performed with aliphatic ketones and various aromatic aldehydes. Especially, (S)methyl 3-mercapto-2-(S)-3-phenyl-2-((S)-pyrrolidine-2-carboxamido)propanamido)propanoate showed good catalytic activities in water at 0 degrees C in the presence of p-nitrobenzoic acid cocatalyst. The enantioselectivities were up to 90.4%, the diastereomeric ratios were up to 97/3 and yields were 99%. The results showed that these organocatalysts were promising organocatalysts for aldol reaction. Besides, this catalyst showed its best catalytic activities in water which is also an important contribution to green chemistry requirements.

Without using chiral sources, the Strecker reaction of achiral hydrogen cyanide, p-tolualdehyde and benzhydrylamine gave enantioenriched L- or D-N-benzhydryl-alpha-( p-tolyl) glycine nitriles with up to > 99% ee in a mixed solvent of water and methanol. Therefore, total spontaneous resolution of alpha-aminonitriles could occur through a prebiotic mechanism of alpha-amino acid synthesis. Moreover, it was demonstrated that the repetition of partial dissolution and crystallization of a suspended conglomerate of aminonitrile under solution-phase racemization could generate the enantiomeric imbalance to afford, in combination with the amplification of chirality, an enantioenriched product in every case. Among the 73 experiments that were carried out, D-and L-enriched isomers occurred 36 and 37 times, respectively. This stochastic behavior, under achiral or racemic starting conditions, meets the requirements of the spontaneous absolute asymmetric Strecker synthesis. The implications of the present results for the origin of chirality of alpha-amino acids are discussed.

Using the easily accessible 2-azaallyl anions as initiators, the one-pot synthesis of well-defined primary amine-ended telechelic polyaziridines (alpha-NH2 PAzs) has been achieved through the ring-opening polymerization (ROP) of N-sulfonyl aziridines followed by hydrolysis of the diphenylketimine moiety (-N=C-Ph-2). The 2-azaallyl anions were synthesized from the reaction of diphenylketimine or N-[aryl-methylene]-alpha-phenylbenzenemethanamine with potassium bis(trimethylsilyl) amide (KHMDS) in situ and used to initiate the ROP of aziridines leading to well-defined alpha-(Ph2C=N)-alpha’-aryl-omega-NH PAzs. Along with the diphenylketimine group (-N=C-Ph-2), aryl functionalities, such as pyridine and triphenylphosphine moieties, can also be incorporated to the chain end. Chain extension has been applied for the synthesis of poly(N-sulfonyl aziridine)-block-poly(epsilon-caprolactone) (PAz-b-PCL) block copolymers by utilization of the primary amine end group as initiating sites for the ROP of epsilon-caprolactone catalyzed by tin 2-ethyl hexanoate (SnOct(2)). Taking advantage of this synthetic approach, core cross-linked multiarm star (CCS) polymers with an outermost shell having amino and triphenylphosphine functionalities have been synthesized via arm-first strategy.

When activated with MAO or MMAO in toluene, these complexes showed relatively low activities but afforded polymers with ultra-high molecular weight (up to 3.30 x 10(6) Da). 1-6/Et2AlCl also showed high activities in ethylene/1-hexene copolymerization at room temperature giving moderate molecular-weight polymers (6.5-11.4 x 10(4) Da) with co-monomer incorporation being of 6.0 similar to 7.8%.
