Effects of RVD-hemopressin (alpha) on feeding and body weight after standard or cafeteria diet in rats

Palatability and variety of foods are major reasons for hedonic eating, and hence for obesity. Hemopressin, a hemoglobin alpha chain-derived peptide, plays antagonist/inverse agonist role on cannabinoid (CB)1 receptors, while RVD-hemopressin(alpha)[RVD-hp(alpha)], a N-terminally extended form of hemopressin, has been reported as an allosteric modulator of CB1 and CB2 receptors. We investigated the effects of 14 daily intraperitoneal injections of RVD-hp(alpha), in Sprague-Dawley rats fed a highly palatable cafeteria-style (CAF) diet (30% fat, 56% carbohydrate, 14% protein; 4.20 kcal/g) compared to standard laboratory chow (STD) food (3.5% fat, 63% carbohydrate, 14% protein, 19.5% other components without caloric value; 3.20 kcal). Food intake, body weight and locomotor activity were recorded throughout the study. Finally, rats were sacrificed and agouti-related peptide (AgRP), neuropeptide Y (NPY), pro-opiomelanocortin (POMC) and cocaine and amphetamine-regulated transcript (CART) and fatty acid amide hydrolase (FAAH) gene expression in the hypothalamus was measured by real-time reverse transcription polymerase chain reaction. We found that CAF diet increased food intake as compared to STD diet. In both STD and CAF diet fed rats, RVD-hp(alpha) treatment inhibited food intake, increased locomotor activity but did not modify body weight. In vehicle injected animals, CAF as compared to STD diet increased AgRP gene expression. RVD-hp(alpha) treatment decreased POMC mRNA levels in both diet groups and lowered the elevated AgRP levels induced by CAF diet. RVD-hp(alpha) treatment plays an anorexigenic role paralleled by increased locomotor activity both in STD and CAF diet fed rats. The inhibition of feeding could be partially mediated by lowering of hypothalamic POMC and AgRP gene expression levels.

Fragmentation pathways of deprotonated amide-sulfonamide CXCR4 inhibitors investigated by ESI-IT-MSn, ESI-Q-TOF-MS/MS and DFT calculations

Amide-sulfonamides provide a potent anti-inflammatory scaffold targeting the CXCR4 receptor. A series of novel amide-sulfonamide derivatives were investigated for their gas-phase fragmentation behaviors using electrospray ionization ion trap mass spectrometry and quadrupole time-of-flight mass spectrometry in negative ion mode. Upon collision-induced dissociation (CID), deprotonated amide-sulfonamides mainly underwent either an elimination of the amine to form the sulfonyl anion and amide anion or a benzoylamide derivative to provide sulfonamide anion bearing respective substituent groups. Based on the characteristic fragment ions and the deuterium-hydrogen exchange experiments, three possible fragmentation mechanisms corresponding to ion-neutral complexes including [sulfonyl anion/amine] complex (INC-1), [sulfonamide anion/benzoylamide derivative] complex (INC-2) and [amide anion/sulfonamide] complex (INC-3), respectively, were proposed. These three ion-neutral complexes might be produced by the cleavages of S-N and C-N bond from the amide-sulfonamides, which generated the sulfonyl anion (Route 1), sulfonamide anion (Route 2) and the amide anion (Route 3). DFT calculations suggested that Route 1, which generated the sulfonyl anion (ion c) is more favorable. In addition, the elimination of SO2 through a three-membered-ring transition state followed by the formation of C-N was observed for all the amide-sulfonamides.

Performance of commercial composite hydrophobic membranes applied for pervaporative reclamation of acetone, butanol, and ethanol from aqueous solutions: Binary mixtures

In this study the efficiency of three various commercial membranes based on poly(octylmethyl siloxane), poly (ether-block-amide), and poly(dimethylsiloxane) polymers were investigated in pervaporative separation of acetone, butanol, and ethanol from aqueous binary solutions (0-5 wt% of organics) at 60 degrees C. The influence of fermentation broth microfiltration on membrane performances in pervaporative removal of ethanol was also investigated. The use of microfiltration improved the effectiveness of the removal of ethanol from the broth comparing with the unfiltered one. Molar ratios of organics to water fluxes and Pervaporative Separation Index (PSI) values were employed to discuss membranes’ performance in removal of organic solvents from binary aqueous mixture. It was found that PDMS based (Pervap4060) membrane shows the best separation efficiency in all tested binary aqueous mixtures. Modelling of the process proved the feasibility of pervaporation process for the removal of acetone, butanol and ethanol from binary and quaternary aqueous mixtures.

Grafting of poly(methacrylic acid-co-acrylamide) film on silicon surface via a simultaneous hydrolysis process

In this paper, we demonstrate a one-pot method to prepare the poly(methacrylic acid-co-acrylamide) film on silicon surface by the simultaneous polymer grafting and hydrolysis of the acrylonitrile. The surface morphology, grafting density, chemical structures, grafting ratio and the hydrolysis process were studied to explore the film growth. The thickness were linearly increasing in the grafting process, which indicated the one-pot grafting process was active and controllable. During the grafting process, cyano groups were totally hydrolysed to amide groups under the catalysis of hydrogen ions. This simple grafting method overcomes the grafting difficulty of acrylamide by diazonium salt, which may be protonated in acidic solution. Besides, this method may also be utilized to graft high-density copolymer films containing polyacrylamide chains onto a range of semiconductor and metal surfaces by diazonium salt chemistry.

Performance improvements of the BNC tubes from unique double-silicone-tube bioreactors by introducing chitosan and heparin for application as small-diameter artificial blood vessels

In order to improve property of bacterial nano-cellulose (BNC) to achieve the requirements of clinical application as small caliber vascular grafts, chitosan (CH) was deposited into the fibril network of the BNC tubes fabricated in unique Double-Silicone-Tube bioreactors. Heparin (Hep) was then chemically grafted into the BNC-based tubes using EDC/NHS crosslinking to improve performance of anticoagulation and endothelialization. Physicochemical and mechanical property, blood compatibility, and cytocompatibility were compared before and after compositing. The results indicated that strength at break was increased but burst pressure decreased slightly after compositing. Performance of the BNC tubes was improved remarkably after introducing chitosan and heparin. The EDC/NHS crosslinking catalyzed both amide bonds and ester bonds formation in the BNC/CH-Hep composites. Three-dimensional surface structure and roughness were firstly obtained and discussed in relation to the hemocompatibility of BNC-based tubes. This work demonstrates the heparinized BNC-based tubes have great potential in application as small-diameter vascular prosthesis.

Structure and optical properties of transparent polyamide 6 containing lithium bromide

We investigated the effects of lithium bromide (LiBr) on the structure and properties of polyamide 6 (PA6). The strong ion-dipole interactions between lithium cations and the amide groups in PA6 greatly increased the glass transition temperature (T-g) and retarded the crystallization rate of PA6. As a result, compression-molded PA6 blends were highly transparent and had high T-g values. The rheological terminal region was obvious in the blends because the ion-dipole interactions weakened at high temperatures. This indicates that the melt processability was barely affected by LiBr. We also evaluated the optical anisotropy of the polymer to determine its suitability as a functional optical film. We found that hot-stretched blend films had large positive orientation birefringence with significantly weak wavelength dispersion, which can be attributed to the enhanced anisotropic polarizability of PA6. Deep purification of seawater using a novel zeolite 3A incorporated polyether-block-amide composite membrane

In this study, a novel hydrophilic-hydrophobic composite membrane was prepared and used in a pervaporative desalination of NaCl-water solution and seawater. For this purpose, polyether block amide (PEBA) was selected as membrane matrix. Zeolite 3A was incorporated to PEBA polymer for facilitating the water permeation through the membrane. The surface morphologies of the pristine and composite membranes were examined by scanning electron microscopy. Thermogravimetric analyses of the pristine and composite membranes were performed by adjusting the zeolite 3A concentration in the PEBA polymer. The effect of the zeolite addition on membrane’s surface hydrophilicity was analyzed using contact angle measurement. Firstly, influences of zeolite content, feed temperature and NaCl concentration on pervaporative desalination performances were performed in the pervaporation of NaCl-water solution. All membranes exhibited excellent performance, and the salt rejection of >99.5% and flux of >2.07 kg m(-2) h(-1) were achieved. Secondly, seawater desalination was performed. Effect of zeolite addition at a given temperature was also investigated. The better salt rejection was obtained as 99.81% accompanied with a very good flux of 4.57 kg m(-2) h(-1) in pervaporative seawater desalination at 40 degrees C using 20 wt.% zeolite 3A incorporated membrane. Chemical cross-linking on gelatin-hyaluronan loaded with hinokitiol for the preparation of guided tissue regeneration hydrogel membranes with antibacterial and biocompatible properties

The mechanical properties and structural stability of hydrogels and their performance in antidegradation can be enhanced by cross-linking them with N-(3-dimethylaminopropyl)-N’-ethylcarbodiimide hydrochloride (EDC). However, residual EDC compromises the biocompatibility of cross-linked hydrogels and the formability of uncross-linked hydrogels. In this study, a facile process for preparing hydrogel regenerative membranes exerting antibacterial effects and containing gelatin/hyaluronic acid (G/HA) through solution casting was proposed. The membranes were cross-linked with EDC (G/HA-Ec-0H) and impregnated with two concentrations of the antibacterial agent of hinokitiol (G/HA-Ec-2H and G/HA-Ec-4H). Amide bonds formed, and the rate of active amino acid fixation was higher than 90%, which was directly proportional to the degree of cross-linking. The G/HA-Ec-2H and G/HA-Ec-4H groups with hinokitiol showed good antibacterial properties. The rate of hydrogel degradation decreased, and the integrity of sample morphology was maintained at more than 80% for over 3 days in the immersion. Then, the hydrogel structures relaxed and disintegrated through a rapid degradation reaction within 24 h. The biocompatibility results showed that low concentrations of hinokitiol did not affect cell viability. Moreover, hydrogel membranes after 14 days of cell incubation showed good cell adhesion and proliferation. In summary, the membrane biostability of the cross-linked gelatin/hyaluronan hydrogels was enhanced by EDC at a biocompatible concentration, and the functionalized group of G/HA-Ec-2H shows potential as a biodegradable material for biocompatible tissue-guarded regeneration membranes with antibacterial properties.

Twofold C-H Activation Enables Synthesis of a Diazacoronene-Type Fluorophore with Near Infrared Emission Through Isosteric Replacement

The synthesis and photophysical properties of a soluble amide-embedded coronene is reported. The key step in this synthesis is the twofold C-H activation of diazaperylene by a rhodium(III)Cp* catalyst. This unprecedented structural motif shows intense fluorescence in the near infrared region with a small Stokes shift and a distinct vibronic structure, which exhibits a slight extent of negative solvatochromism. Comparison of this compound with some relevant compounds revealed the importance of the amide incorporation in the peripheral concave region including an angular position to retain high aromaticity reflecting that of parent coronene. Treatment of this compound with Lewis acid B(C6F5)(3) formed a bis-adduct, which exhibited enhanced aromaticity as a consequence of the increased double bond character of the amide C-N bonds.
