Thermal hydrolysis is a common pre-treatment, used before anaerobic digestion processes, to enhance the hydrolysis rate. However, formation of inhibitory compounds and the increase of liquid fraction colour have been identified as potential drawbacks. This study was oriented to study the methane production from simple substrates, subjected to thermal hydrolysis. A mixture of glycine and glucose at different concentrations was prepared, at a ratio similar to proteins and carbohydrates found in activated sludge. Two temperatures were tested. At 120 degrees C a decrease on biogas production rate was observed. On the other hand, at 165 degrees C generation of recalcitrant material was observed, causing a decrease in methane potential and COD degradation, when a mixture of glycine and glucose was used as substrate. This was atributed to the formation of recalcitrant compounds via Maillard reaction, hyphothesis supported by FTIR-ATR, which indicated the formation of amide II Bonds.

Some Eastern Asian countries deem pufferfish – especially its muscle – a culinary delight. Herein, molecular mass fingerprinting of soups prepared by Takifugu fiavidus, Takifugu obscurus and Takifugu rubripes was established via matrix-assisted laser desorption/ionization – mass spectrometry (MALDI MS). Soup samples were directly analyzed by MALDI MS to collect mass spectra within 0-700 Da in a quick way, followed by principal component analysis to distinguish the different soups and to find out the distinctive compounds among the soups. High performance liquid chromatography – tandem MS (HPLC-MS/MS) was applied to identify the compounds. Nineteen compounds were identified from the HPLC-MS/MS data by using METLIN database. Through literature mining, we found that these compounds are closely related to the flavor, nutrition, and safety of pufferfish soups. This method can also be used as a facile way to distinguish between different pufferfish fillets when morphological characters have been damaged or destroyed.

Oxetanes have been increasingly used as stable motifs in medicinal chemistry as well as versatile synthetic intermediates. Herein, an intramolecular ring opening of oxetane carboxamides with mild nucleophiles, such as nitrogen heterocycles, is presented. The reaction proceeds under metal-free basic conditions which is highly unusual in ring opening reactions of oxetanes. Amide formation and oxetane ring opening/cyclization in a one-pot approach affords high levels of molecular complexity in a single step from simple, readily available substrates.

The reduction of ketones by SmI2-water has long been thought to proceed through a reversible initial electron transfer with the formation of organosamarium intermediates in a follow-up step. Kinetic experiments on the reduction of two model ketones and structurally similar ketones with a pendant alkene are shown to be consistent with a rate-limiting reduction by SmI2-water through a proton-coupled electron-transfer (PCET). Literature values for the rates of radical cyclizations and reduction of radicals by SmI2 and thermochemical data for radical reduction by SmI2-water further support a rate-limiting initial step for ketone reductions. These data suggest that discrete organosamarium species may not be intermediates in ketone reductions by SmI2-water.

We report a novel approach toward the catalytic hydrogenation of CO2 to methanol performed in the gas-solid phase using multinuclear iridium complexes at low temperature (30-80 degrees C). Although homogeneous CO(2 )hydrogenation in water catalyzed by amide-based iridium catalysts provided only a negligible amount of methanol, the combination of a multinuclear catalyst and gas-solid phase reaction conditions led to the effective production of methanol from CO2. The catalytic activities of the multinuclear catalyst were dependent on the relative configuration of each active species. Conveniently, methanol obtained from the gas phase could be easily isolated from the catalyst without contamination with CO, CH4, or formic acid (FA). The catalyst can be recycled in a batchwise manner via gas release and filling. A final turnover number of 113 was obtained upon reusing the catalyst at 60 degrees C and 4 MPa of H-2/CO2 (3:1). The high reactivity of this system has been attributed to hydride complex formation upon exposure to H-2 gas, suppression of the liberation of FA under gas-solid phase reaction conditions, and intramolecular multiple hydride transfer to CO2 by the multinuclear catalyst.

This investigation deals with the design and synthesis of new derivatives of pyrrole consisting of modifying atoms of chlorine, amide, and 1,3-oxazole fragments. These compounds can be interesting in the context of research of new antimicrobial agents. Ethyl 5-chloro-4-formyl-1H-pyrrole-3-carboxylates were used as a key substrate for further transformation into target compounds. This process was realized as a direct transformation of an aldehyde fragment into a 1,3-oxazole cycle by van Leusen’s reaction followed by hydrolysis of an ester group, which finally converted a reactant into the corresponding pyrrole-3-carboxylic acid. This acid has been found to be an efficient construction block for the further development of antimicrobial agents. The preparative potential of these compounds has been verified by way of their transformation into a series of carbamides through consecutive reactions with thionyl chloride and alkyl-, aryl, and heterylamines under mild reaction conditions. According to bio screening results, the following two compounds have been chosen as those exhibiting a high anti-staphylococcus activity: 1-butyl-5-chloro-2-methyl-4-(1,3-oxazol-5-yl)-N-[(1,3-thiazol-2-yl]-1H-pyrrole-3-carboxamide and 1-butyl-5-chloro-N-[(3-dimethylaminosulfonyl)phenyl]-2-methyl-4-(1,3-oxazol-5-yl)-1H-pyrrole-3-carboxamide (MIC = 7.8 mu g/ml), while another one – 5-chloro-N-(4-chlorophenyl)-4-(1,3-oxazol-5-yl)-2-phenyl-1-propyl-1H-pyrrole-3-carboxamide was selected as a compound exhibiting high antifungal activity (MIC = 7.8 mu g/ml) against the reference strains Candida albicans ATCC 885/653 and Aspergillus niger K9.

Mixtures of amino acids with hydroxy acids allow for the formation of peptide bonds in a plausible prebiotic scenario via ester bond formation followed by ester amide exchange. Here, we investigate the ability of the ester-mediated reaction pathway to form even longer polymers with peptide backbones based on the specific details of the reaction protocol. Fresh monomers were fed to the polymer/monomer mixture periodically by an automated day-night machine that was designed to simulate wet-dry cycles that would have been common on the prebiotic Earth. Quantitative analysis of peptide bond formation in the complex oligomer mixture was enabled by a simple hydrolysis treatment. In the ester mediated peptide elongation process, new monomers add to one end of the chain step-by-step without termination. The feed composition (hydroxy acids and/or amino acids) was found to determine the final oligomer distribution. Production of longer oligomers enriched in peptide bonds was more efficient when only amino acids were fed because of a smaller number of active oligomer chains. These results reveal a process for synthesizing longer depsipeptides and/or peptides that could form secondary structures, and possibly functional polymers.

Discovered over a century ago, Beckmann rearrangement is still today fully compliant with all the green chemistry principles and consistent with the key aspects of sustainable development. Herein, we report on a sustainable mechanochemical procedure allowing the design of new amide frameworks via an eco-efficient cut-and-paste process of C-C and C-N bonds on the oxime backbone. We combined inexpensive and readily available reagents, such as p-tosyl imidazole (p-Ts-Im) and oxalic acid, to prepare smoothly and in good to high yields a library of structurally different amides, including value-added marketed compounds such as epsilon-caprolactam and the active pharmaceutical ingredient (API) paracetamol. This solvent-free mechanochemical procedure has also been optimized and successfully extended to several ketones serving as oxime precursors.

The synthesis of 2,4-disubstituted quinazolines by a palladium-catalyzed reaction of arylboronic acids with N-(2-cyanoaryl)benzamides has been developed with moderate to excellent yields. The method shows good functional group tolerance. In particular, halogen and hydroxyl substituents, which are amenable for further synthetic elaborations, are well tolerated. Moreover, the present synthetic route could be readily scaled up to gram quantity without difficulty. The mechanism possibly involves nucleophilic addition to the nitrile function, forming an imine intermediate followed by an intramolecular addition to the amide and dehydration to the quinazoline ring.

In this article, a novel series of (E)-3-(3,4,5-trimethoxyphenyl)acrylic acid (TMCA) amide derivatives 1-18 were designed and synthesized by a facile and one-pot step, which were achieved with good yields using 1-hydroxybenzotriazole (HOBT) and 1-(3-Dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDCI) as activation system. All the synthesized derivatives were biologically evaluated for their anticonvulsant, sedative activity and neurotoxicity using the maximal electroshock (MES) model, sc-pentylenetetrazol (PTZ) model, pentobarbital sodium-induced sleeping model, and locomotor activity tests, respectively. Among them, compounds 4, 9 and 16 exhibited good anticonvulsant activity in primary evaluation. Furthermore, compound 4 is the most effective anticonvulsant and sedative agent in subsequent tests, while the low threshold of toxicity of compound 4 is vigilant. Compounds 9 and 16 also performed significantly anticonvulsant activity in subsequent tests with weak toxicity. The molecular modeling experiments also predicted good binding interactions of the obtained active molecules with the GABA transferas. Therefore, it could be concluded that the synthesized derivatives 4, 9 and 16 would represent useful lead compounds for further investigation in the development of anticonvulsant and sedative agents.
