To explore anticancer and antioxidant agents with improved potency, we synthesized a series of amide linked 1,4-disubstituted 1,2,3-triazoles through click chemistry approach. Molecular docking studies of compounds 7e and 7f was carried out in active site of human epidermal growth factor receptor 2 revealed high binding affinities and within toxicity limits. The experimental results were in good agreement with docking studies. Scarce knowledge on the behavior of vitamins in aqueous solutions in the presence of additives is often a limiting factor for industrial applications such as process design and optimization. Knowing the pH-solubility profiles of vitamins is fundamental for understanding and controlling their behavior in aqueous solutions. In the present work, pH-dependent solubilities of the vitamins ascorbic acid (VC), riboflavin (VB2), nicotinic acid (VB3(acid)), folic acid (VB9), and cyanocobalamin (VB12) were measured at T = 298.15 K and p = 1 bar. These results were compared to the pH-solubility profiles obtained with modified Henderson-Hasselbalch equations using pK(a) values from the literature. Further, the solubilities of poorly soluble VB2, VB9, and VB12 were increased by the addition of covitamins VC, VB3(acid), and nicotinamide (VB3(acid)). As observed, VB3(amide )increases the vitamin solubility much stronger than VC and VB3(acid). These covitamins are called hydrotropes in several works in the literature, and they increase the solubility of other vitamins by manipulating the pH of the saturated solutions and by molecular cross-interactions. The interplay between both pH and cross-interactions depends strongly on the kind and concentration of covitamin. At low concentrations, VC and VB3(amide) (<0.2 m) increased solubility by pH change. At higher concentrations of VC and VB3(amide) added, mainly cross-interactions between vitamin and covitamin determine the strength of solubility increase. To separate these effects and to further reduce experimental effort, electrolyte perturbed-chain statistical association fluid theory was used to predict vitamin solubility. The pH-solubility profiles and the solubilities of vitamins in water at T = 298.15 K and p = 1 bar upon addition of covitamins were predicted with reasonable accuracy. This success resulted from accounting for different charged and neutral vitamin species according to the pH and from considering explicitly the vitamin- water and vitamin-covitamin interactions. It could be shown that hydrotropic solubilization of a vitamin is the increase of vitamin solubility caused by pH shift and by cross-interactions between the saturated species of a vitamin and the added covitamin. After 10 min subcritical hydrothermal treatment with microalgal solid concentration of 5% at 245 degrees C and esterification reactions (100 degrees C)with methanol and H2SO4 catalysts, the solvent-free lipids extraction efficiency was improved from 69.9% (260 degrees C) to 95.0 wt% without losing the valuable unsaturated fatty acids in microalgae cells. The resulted main compositions of C16:0, C16:1 and C20:5 in microalgal biodiesel reached the highest of 22.7%, 24.1% and 16.2% (based on total lipids in microalgae), much higher than those lipids obtained from 260 degrees C treatment (C20:5 = 6.1%). Standard methods of protein aggregation analysis- Thioflavin T (ThT) fluorescence, atomic force microscopy (AFM), and 8-anilinonaphthalene-1-sulphonic acid (ANS) fluorescence were applied and compared to FTIR spectroscopy data. Results show the great potential of FTIR for both, qualitative and quantitative monitoring of oligomer formation based on the secondary structure changes. While oligomer intermediates do not induce significant changes in ThT fluorescence, their secondary structure changes were very prominent. Normalization of specific Amide I region peak intensities by using Amide II peak intensity as an internal standard provides an opportunity to use FTIR spectroscopy for both qualitative and quantitative analysis of biological samples and detection of potentially toxic oligomers, as well as for screening of efficiency of fibrillation procedures.

Following a previous study that focused on the organic signatures of fireplaces fueled with wood and/or bones, this study targeted the completion of an experimental reference database through the addition of a wood-fueled fireplace dedicated to the cooking of meat. Different sedimentary features of this experimental fireplace were visually identified (e.g. darkening, reddening), sampled, and subjected to geochemical analysis. Corg and N contents were quantified, samples were extracted with organic solvents and analyzed through GC-MS and bulk organic matter was characterized through py(TMAH)-GC-MS. Five different samples were studied and compared with a control sample, representative of the local background. A significant increase of Corg and N contents was measured for the three samples presenting darkened or charred caracteristics. The meat-cooking fireplace seems to be characterized by the strong contribution of nitrogen, which was visible in elementary analyses as well as in the molecular composition of solvent extracts, and bulk organic matter. More specifically, compounds containing nitrile functions, amides, N-heterocyclic and N-aromatic compounds could be detected in solvent extracts. Amines, amides, N-heterocyclic and N-aromatic compounds could be identified in py(TMAH)-GC-MS. Some of these compounds present a relative stability in soils and could therefore aid in our comprehension and functional interpretations of archaeological fireplaces, and may, more particularly, make it possible to highlight the use of fireplaces for the cooking of meat.
