Altered activity of fatty acid amide hydrolase (FAAH), an enzyme of the endocannabinoid system, has been implicated in several neuropsychiatric disorders, including major depressive disorder (MDD). It is speculated that increased brain FAAH expression is correlated with increased depressive symptoms. The aim of this scoping review was to establish the role of FAAH expression in animal models of depression to determine the translational potential of targeting FAAH in clinical studies. A literature search employing multiple databases was performed; all original articles that assessed FAAH expression in animal models of depression were considered. Of the 216 articles that were screened for eligibility, 24 articles met inclusion criteria and were included in this review. Three key findings emerged: (1) FAAH expression is significantly increased in depressive-like phenotypes; (2) genetic knockout or pharmacological inhibition of FAAH effectively reduces depressive-like behavior, with a dose-dependent effect; and (3) differences in FAAH expression in depressive-like phenotypes were largely localized to animal prefrontal cortex, hippocampus and striatum. We conclude, based on the animal literature, that a positive relationship can be established between brain FAAH level and expression of depressive symptoms. In summary, we suggest that FAAH is a tractable target for developing novel pharmacotherapies for MDD.

Fatty acids-assisted superparamagnetic maghemite (gamma-Fe2O3) NPs was biologically synthesized using extract of polyherbal drug Liv52 (L52E). The synthesized NPs were used to investigate their antimicrobial, antibiofilm activity against P. aeruginosa, MRSA and C. albicans and anticancer activity on colon cancer cells (HCT-116) for biomedical applications. Further, molecular docking study was performed to explore the interaction of Fe2O3 NPs with major cell wall components i.e., peptidoglycan and mannoproteins. The docking studies revealed that Fe2O3 interacted efficiently with peptidoglycan and mannoproteins and Fe2O3 get accommodated into catalytic cleft of mannoprotein. Due to magnetic property, the biological activity of gamma-Fe2O3 can be further enhanced by applying external magnetic field alone or in amalgamation with other therapeutics drugs. Further, peripheral amine groups were grafted with succinic anhydride to achieve carboxylic acid terminated dendrimer. The resulting nanomaterial was confirmed by FTIR, XRD, VSM, TGA, HRTEM and conductometry methods. In order to emphasize the structural efficacy of polyamine dendrimer, the metal ion uptake behavior has been discussed for the representative cations such as Pb2+, Cu2+, Cr6+, Ca2+ and Ni2+ under the environmental concern. The aims of this study are to isolate and characterize acid soluble collagen (ASC) and pepsin soluble collagen (PSC) extracted from silver catfish (Pangasius sp.) skin. Isolated ASC and PSC collagen were characterized in terms of chemical composition (moisture, protein, fat and ash content), protein concentration, functional group, solubility, and morphological properties as compared to commercial collagen. Yields of ASC and PSC were 4.27% and 2.27%, respectively. The chemical compositions of raw skin were 34.64%, 2.81%, 3.68%, and 0.31%, while the chemical compositions of ASC and PSC were 94.21%, 3.48%, 0.81%, 59.15%, and 88.25%, 3.46%, 0.92%, and 29.24%, for moisture, protein, fat, and ash, respectively. ASC and PSC had protein concentrations of 2.27 mg/mL and 2.70 mg/mL, respectively. Functional group analysis revealed that both isolated collagens exhibited Amide A, II and III as a fingerprint for collagen structure. The highest solubility was found at pH 4 for ASC, pH 1 for PSC, and pH 5 for commercial collagen. The morphology of the isolated collagens was porous and they contained fibril. In conclusion, the characteristics of the isolated ASC and PSC from silver catfish (Pangasius sp.) skin indicate that value-added collagen can be produced from the alternative source of freshwater fish. Inhibitors of human neuraminidase enzymes (NEU) are recognized as important tools for the study of the biological functions of NEU and will be potent tools for elucidating the role of these enzymes in regulating the repertoire of cellular glycans. Here we report the discovery of selective inhibitors of the human neuraminidase 1 (NEU1) and neuraminidase 2 (NEU2) enzymes with exceptional potency. A library of modified 2-deoxy-2,3-didehydro-N-acetylneuraminic acid (DANA) analogues, with variability in the C5- or C9-position, were synthesized and evaluated against four human neuraminidase isoenyzmes (NEU1-4). Hydrophobic groups with an amide linker at the C5 and C9 positions were well accommodated by NEU1, and a hexanamido group was found to give the best potency at both positions. While the CS-hexanamido-C9-hexanamido-DANA analogue did not show synergistic improvements for combined modification, an extended alkylamide at an individual position combined with a smaller group at the second gave increased potency. The best NEU1 inhibitor identified was a C5-hexanamido-C9-acetamido-DANA that had a K-i of 53 +/- 5 nM and 340-fold selectivity over other isoenzymes. Additionally, we demonstrated that C5-modifications combined with a C4-guandino group provided the most potent NEU2 inhibitor reported, with a K-i of 1.3 +/- 0.2 mu M and 7-fold selectivity over other NEU isoenzymes.
