Computational studies were also performed using DFT method at B3LYP/6-311G(d,p) level of theory. Vibrational modes and chem. shifts were calculated and compared with the exptl. data. The genes tetA, tetX and sulI were related to the most diverse bacteria that were most able to proliferate during heap composting. In this study, through the use of a conformationally-constrained design strategy, a series of thieno[3,2-d]pyrimidines were synthesized as potent JAK3 inhibitors for the treatment of IPF. Among them, the most potent JAK3 inhibitor, namely 8e (IC50 = 1.38 nM), significantly reduced the degree of airsacculitis and fibrosis according to hematoxylin-eosin (HE) staining assay for the lung tissue in the bleomycin (BLM)-induced pulmonary fibrosis mouse model. The clear reduction of the lung collagen deposition by the determination of Masson and hydroxyproline (HYP) content also demonstrated its efficacy in the treatment of fibrosis. In addition, 8e also reduced the expression of the inflammatory markers IL-6, IL-17A, TNF-α and malondialdehyde (MDA) in lung tissue, which indicated its higher anti-inflammatory activity compared with that of the reference agents (nintedanib and gefitinib). Degradation of seven relevant pharmaceuticals with different chem. structures and properties: acetaminophen (ACE), cloxacillin (CXL), diclofenac (DCF), naproxen (NPX), piroxicam (PXC), sulfacetamide (SAM) and cefadroxil (CDX), in distilled water and mineral water by ultrasound was studied herein. Firstly, proper conditions of frequency and acoustic power were determined based on the degradation ability of the system and the accumulation of sonogenerated hydrogen peroxide (24.4 W and 375 kHz were found as the suitable conditions for the sonochem. treatment of the pharmaceuticals). Under such conditions, the pharmaceuticals degradation order in distilled water was: PXC > DCF ∼ NPX > CXL > ACE > SAM > CDX. In fact, the initial degradation rate showed a good correlation with the Log P parameter, most hydrophobic compounds were eliminated faster than the hydrophilic ones. Interestingly, in mineral water, the degradation of those hydrophilic compounds (i.e., ACE, SAM and CDX) was accelerated, which was attributed to the presence of bicarbonate ions. Afterwards, mineral water containing six different initial concentrations (i.e., 0.331, 0.662, 3.31, 16.55, 33.1, and 331μM) of selected pharmaceuticals was sonicated, the lowest concentration (0.331μM) always gave the highest degradation of the pollutants. This result highlights the great ability of the sonochem. process to treat bicarbonate-rich waters containing pollutants at trace levels, as pharmaceuticals. Finally, the addition of ferrous ions to the sonochem. system to generate a sono-Fenton process resulted in an acceleration of degradation in distilled water but not in mineral water. This was attributed to the scavenging of sonogenerated HO• by bicarbonate anion, which decreases H2O2 accumulation, thus limiting the Fenton reaction. The present study firstly aimed at developing a multi-residue method to identify and quantify 38 veterinary antibiotics (belonging to five different classes) not only for raw swine wastewater but also for wastewater differently treated by different units. The proposed method is based on a solid-phase extraction procedure and ultra high performance liquid chromatog. with mass spectrometry. For sample preparation, the optimal loading sample volume was selected as 50 mL, the pH of which was adjusted to approx. 3.0 using formic acid. Then 0.1 g/L EDTA disodium salt was added. The recovery rates for different types of wastewaters were in the range of 35.94-124.51% and the relative standard deviations were in the range of 0.36-14.62%. All the matrix standard curves exhibited high linearity (0.9956-0.9999). The matrix effects for the target antibiotics ranged from -61.73 to +148.75%. To ensure the practicality of the method, we performed the detection of the actually added concentration to determine method detection limits and quantitation limits. The quantitation limits of most of the target antibiotics were 0.04μg/L, except for spiramycin (0.1μg/L) and roxithromycin (0.2μg/L). This optimized and validated method was applied to analyze antibiotic residues in swine water samples from four swine farms.
