Salt effects on the solubility of uncharged polymers in aqueous solutions are usually dominated by anions, while the role of the cation with which they are paired is often ignored. In this study, we examine the influence of three aqueous metal iodide salt solutions (LiI, NaL and CsI) on the phase transition temperature of poly(N-isopropylacrylamide) (PNIPAM) by measuring the turbidity change of the solutions. Weakly hydrated anions, such as iodide, are known to interact with the polymer and thereby lead to salting-in behavior at low salt concentration followed by salting-out behavior at higher salt concentration. When varying the cation type, an unexpected salting-out trend is observed at higher salt concentrations, Cs+ > Na+ > Li+. Using molecular dynamics simulations, it is demonstrated that this originates from contact ion pair formation in the bulk solution, which introduces a competition for iodide ions between the polymer and cations. The weakly hydrated cation, Cs+, forms contact ion pairs with I- in the bulk solution, leading to depletion of CsI from the polymer-water interface. Microscopically, this is correlated with the repulsion of iodide ions from the amide moiety.

Polyacrylonitrile nanoparticles grafted on ethylene diamine functionalized partially reduced graphene oxide (PAN-PRGO) was prepared via in situ emulsion polymerization and was further modified to contain amidoxime, amdinoethylene diamine, and carboxylic groups on the surface of the graphene nanosheets via partial hydrolysis of the nitrile groups on the polymer chains of the composite using (4% NaOH, 20 min) (HPAN-PRGO). The results revealed that polyacrylonitrile nanoparticles were grafted on the surface of the aminated graphene oxide nanosheets via the reaction between the free amino groups of the ethylene diamine modified graphene oxide nanosheets and nitrile groups of acylonitrile (AN). The obtained HPAN-PRGO composite was evaluated for its chelating property with Hg(II) ions. The effect of initial pH, initial concentration of the Hg(II), adsorbent dose, and contact time on the extraction of Hg(II) ions using HPAN-PRGO were investigated. The adsorption experiments indicated that HPAN-PRGO exhibits higher affinity toward Hg(II). The maximum uptake capacity for the extraction of Hg(II) ions on HPAN-PRGO was 324.0 mg/g at pH 5. The HPAN-PRGO shows a 100% removal of Hg(II) at concentrations up to 50 ppm, and the adsorption is exceptionally rapid showing more than 80.0% removal within 15 min and 100.0% of q(e) within 1.5 h at 800 ppm concentration. The Langmuir isotherm model and pseudo-second-order kinetic model have showed good fitness with the practical data. The XPS analysis of HPAN-PRGO before and after adsorption revealed the chelation adsorption mechanism between mercury and amine, amide, amidoxime, and carboxylic groups. After six adsorption-desorption cycles, the HPAN-PRGO could retain more than 90.0% of its original adsorption capacity. These results confirmed that HPAN-PRGO has exceptional performance for the removal of Hg(II) from wastewater. [GRAPHICS] .

Tanneries are the primary source of toluene pollution in the environment and toluene due to its hazardous effects has been categorized as persistent organic pollutant. Present study was initiated to trace out metabolic fingerprints of three toluene-degrading bacteria isolated from tannery effluents of Southern Punjab. Using selective enrichment and serial dilution methods followed by biochemical, molecular and antibiotic resistance analysis, isolated bacteria were subjected to metabolomics analysis. GC-MS/LC-MS analysis of bacterial metabolites helped to identify toluene transformation products and underlying pathways. Three toluene-metabolizing bacteria identified as Bacillus paralicheniformis strain KJ-16 (IUBT4 and IUBT24) and Brevibacillus agri strain NBRC 15538 (IUBT19) were found tolerant to toluene and capable of degrading toluene. Toluene-degrading potential of these isolates was detected to be IUBT4 (10.35 +/- 0.084 mg/h), IUBT19 (14.07 +/- 3.14 mg/h) and IUBT24 (11.1 +/- 0.282 mg/h). Results of GC-MS analysis revealed that biotransformation of toluene is accomplished not only through known metabolic routes such as toluene 3-monooxygenase (T3MO), toluene 2-monooxygenase (T2MO), toluene 4-monooxygenase (T4MO), toluene methyl monooxygenase (TOL), toluene dioxygenase (Tod), meta-and orthoring fission pathways. But additionally, confirmed existence of a unique metabolic pathway that involved conversion of toluene into intermediates such as cyclohexene, cyclohexane, cyclohexanone and cyclohexanol. LC-MS analysis indicated the presence of fatty acid amides, stigmine, emmotin A and 2, 2-dinitropropanol in supernatants of bacterial cultures. As the isolated bacteria transformed toluene into relatively less toxic molecules and thus can be preferably exploited for the eco-friendly remediation of toluene.

The lengths of Si-F-ax. comprise 1.686, 1.661 and 1.632 angstrom respectively. QTAIM analysis was used to estimate the energy of co-ordination bond C=O -> Si in these amides. Post-translational modification of the adeno-associated virus capsids is a poorly understood factor in the development of these viral vectors into pharmaceutical products. Here we report the extensive capsid deamidation of adeno-associated virus serotype 8 and seven other diverse adeno-associated virus serotypes, with supporting evidence from structural, biochemical, and mass spectrometry approaches. The extent of deamidation at each site depended on the vector’s age and multiple primary-sequence and three-dimensional structural factors. However, the extent of deamidation was largely independent of the vector recovery and purification conditions. We demonstrate the potential for deamidation to impact transduction activity and, moreover, correlate an early time point loss in vector activity to rapidly progressing spontaneous deamidation at several adeno-associated virus 8 asparagines. We explore mutational strategies that stabilize side-chain amides, improving vector transduction and reducing the lot-to-lot molecular variability that presents a key concern in biologics manufacturing. This study illuminates a previously unknown aspect of adeno-associated virus capsid heterogeneity and highlights its importance in the development of these vectors for gene therapy.

Here, we present a thorough theoretical study of the adsorption of acrolein (ACO), acrylonitrile (ACN), and acrylamide (ACA) on Cu(100) surface. For this purpose, we have used the density functional theory, imposing periodic boundary conditions to have a correct description of the electronic band structure of the metal and including dispersion forces through two different schemes: the D2 method of Grimme and the vdW-DF. We have found several adsorption geometries. In all of them, the vinyl group together with the amide (in ACA), ciano (in ACN), and carbonyl (in ACO) groups, is highly involved. The highest adsorption energy is found for acrylamide, followed by acrolein and the lowest for acrylonitrile (depending on the level of theory employed 1.0, and 0.9 eV, respectively). We show that a strong coupling between the pi electronic system (both occupied and virtual orbitals) and the electronic levels of the metal is mainly responsible of the chemisorption. As a consequence, electronic density is transferred from the surface to the molecule, whose carbon atoms acquire a partial sp(3) hybridization. Lone-pair orbitals of the cyano, amide, and carbonyl groups also play a role in the interaction. The simulations and following analysis allow to disentangle the nature of the interaction, which can be explained on the basis of a simple chemical picture: donation from the occupied lone pair and pi orbitals of the molecule to the surface and backdonation from the surface to the pi* orbital of the molecule (pi-backbonding).

Calixarenes, with potential functionalization on the upper and lower rim, have been explored in recent years for the design and construction of anticancer agents in the field of drugs and pharmaceuticals. Herein, optimization of bis [N-(2-hydroxyethyl) aminocarbonylmethoxyl substituted calix [4] arene (CLX-4) using structure-based drug design and traditional medicinal chemistry led to the discovery of series of calix [4]arene carbonyl amide derivatives 5a-5t. Evaluation of the cytotoxicity of 5a-5t employing MTT assay in MCF-7, MDA-MB-231 (human breast cancer cells), HT29 (human colon carcinoma cells), HepG2 (human hepatocellular carcinoma cells), A549 (human lung adenocarcinoma cells) and HUVEC (Human Umbilical Vein Endothelial) cells demonstrated that the most promising compound 5h displayed the most superior inhibitory effect against A549 and MDA-MB-231 cells, which were 3.2 times and 6.8 times of CLX-4, respectively. In addition, the cell inhibition rate (at 10 mu M) against normal HUVEC cells in vitro was only 9.6%, indicating the safty of compound 5h. Moreover, compound 5h could inhibit the migration of MDA-MB-231 cell in wound healing assay. Further mechanism studies significantly indicated that compound 5h could block MDA-MB-231 cell cycle arrest in G0/G1 phase by down regulating cyclin D1 and CDK4, and induce apoptosis by up-regulation of Bax, down-regulation of Caspase-3, PARP and Bcl-2 proteins, resulting in the reduction of DNA synthesis and cell division arrest. This paper investigates the correlation between fatigue performance and static tensile behavior of fiber reinforced rubber materials and presents change of fiber reinforced effect during fatigue process. A rough aramid fiber (AF) surface, which is helpful for combining AF and rubber matrix, was achieved by etching AF in NaOH/CH3CH2OH solution via the hydrolysis of amide bonds on the fiber. Then, the treated fiber was coated with an epoxy-functionalized silane (gamma-propyltrimethoxysilane, KH560) and was used to prepare AF/carbon black/styrene-butadiene rubber (AF/CB/SBR) composites. The results showed that the interfacial adhesion between the complex-modified AF and the rubber matrix was further enhanced and the mechanical properties of the composite were further improved compared with those of the composite which was prepared by unhydrolyzed AF coated with KH560. The hysteresis reinforcing effect of the fiber was found after 30,000 cycles of strain-controlled fatigue, and the reinforcing effect of AF was reduced with increasing average fatigue strain. Moreover, the relative debonding energy (RDE) was calculated to evaluate the reinforcing effect of the fiber, and the RDE value sharply decreased after 30,000 cycles of strain-controlled fatigue. When the AF was treated with the NaOH/CH3CH2OH solution and then coated with KH560, the rubber composite had a high RDE value, and the fatigue life of the composite under the stress-control condition was increased approximately 4 times. The results from the characterization of the fracture appearance after 30,000 cycles of fatigue showed that the composite with the complex-modified AF could maintain better interface bonding during the fatigue process.

Pulmonary arterial hypertension (PAH) is a chronic cardiovascular disease that displays inflammatory components, which contributes to the difficulty of adequate treatment with the available therapeutic arsenal. Using X-ray powder diffraction, calorimetric techniques, and molecular modeling, we clearly indicate the presence of a preferred synperiplanar conformation at the amide function, which is fixed by an intramolecular 1,5-N center dot center dot center dot S or alpha-hole intramolecular interaction. Moreover, the evaluation of LASSBio-1835 (4) as a PDE4 inhibitor and as an A(2A) agonist confirms it presents a more balanced dual profile, being considered a promising prototype for the treatment of PAH.
