Owing to its high sensitivity, surface-enhanced Raman spectroscopy (SERS) is widely used in biosensing as an efficient optical transduction technique. In this work, we report the elaboration of a novel SERS-powered aptasensing substrate for the ultra-sensitive and selective detection of prostate specific antigen (PSA). The transducing SERS substrate is formed by vertically aligned silicon nanowires (SiNWs) decorated with silver nanoparticules (AgNPs). The Ni2+ complexes with hexadentate ligands containing two 6-methylpicolinamide groups linked by ethane-1,2-diamine (dedpam) or cyclohexane-1,2-diamine (chxdedpam) spacers were investigated as potential contrast agents in magnetic resonance imaging (MRI). However, the amide derivative [Ni(chxdedpam)](2+) was found to be considerably more inert with respect to proton-assisted dissociation than the carboxylate derivative [Ni(chxdedpa)]. A detailed H-1 NMR and DFT study was conducted to assign the H-1 NMR spectra of the [Ni(chxdedpa)] and [Ni(chxdedpam)](2+) complexes. The observed H-1 NMR paramagnetic shifts were found to be dominated by the Fermi contact contribution. The amide resonances of [Ni(chxdedpam)](2+) at 91.5 and 22.2 ppm were found to provide a sizeable chemical exchange saturation transfer effect, paving the way for the development of NiCEST agents based on these rigid non-macrocyclic platforms.

The increasing emergence of multi-drug resistant bacteria is a serious threat to public health worldwide. Antimicrobial peptides have attracted attention as potential antibiotics since they are present in all multicellular organisms and act as a first line of defence against invading pathogens. We have previously identified a small all-D antimicrobial octapeptide amide kk(1-nal)fk(1-nal)k(nle)-NH2 (D2D) with promising antimicrobial activity. In this work, we have performed a structure-activity relationship study of D2D based on 36 analogues aimed at discovering which elements are important for antimicrobial activity and toxicity. These modifications include an alanine scan, probing variation of hydrophobicity at lys(5) and lys(7), manipulation of amphipathicity, N-and C-termini deletions and lys-arg substitutions. We found that the hydrophobic residues in position 3 (1-nal), 4 (phe), 6 (1-nal) and 8 (nle) are important for antimicrobial activity and to a lesser extent cationic lysine residues in position 1, 2, 5 and 7. Our best analogue 5, showed MICs of 4 mu g/mL against A. baumannii, E. coli, P. aeruginosa and S. aureus with a hemolytic activity of 47% against red blood cells. Furthermore, compound 5 kills bacteria in a concentration-dependent manner as shown by time-kill kinetics. Circular dichroism (CD) spectra of D2D and compounds 1-8 showed that they likely fold into alpha-helical secondary structure. Small angle x-ray scattering (SAXS) experiments showed that a random unstructured polymer-like chains model could explain D2D and compounds 1, 3, 4, 6 and 8. Solution structure of compound 5 can be described with a nanotube structure model, compound 7 can be described with a filament-like structure model, while compound 2 can be described with both models. Lipid interaction probed by small angle X-ray scattering (SAXS) showed that a higher amount of compound 5 (similar to 50-60%) inserts into the bilayer compared to D2D (similar to 30-50%). D2D still remains the lead compound, however compound 5 is an interesting antimicrobial peptide for further investigations due to its nanotube structure and minor improvement to antimicrobial activity compared to D2D.

The in vitro antibacterial activities of the highest binding affinity docked compounds were tested against E. coli, Staphylococcus aureus, and Salmonella spp. Majority of the tested compounds showed effective positive results against E. coli, while they were almost inactive against Staphylococcus aureus and Salmonella spp. The in vitro cytotoxic activities of the highest binding affinity-docked compounds were tested against the human liver carcinoma cell line (HepG2). Some compounds showed potent cytotoxic activity with low IC50 values, especially for 3 (6.525 mu M) and 13a (10.97 mu M) than that for standard drug doxorubicin (2.06 mu M).

In this study, we report the structure-activity relationship profiling of a 6-amino[1,2,5]oxadiazolo[3,4-b]pyrazin-5-ol core, which utilizes the hydroxy moiety as the proton transporter across the mitochondrial inner membrane. We demonstrate that a wide array of substituents is tolerated with this novel scaffold that increased cellular metabolic rates in vitro using changes in oxygen consumption rate as a readout. In particular, compound SHS4121705 (12i) displayed an EC50 of 4.3 mu M in L6 myoblast cells and excellent oral bioavailability and liver exposure in mice. In the STAM mouse model of NASH, administration of 12i at 25 mg kg(-1) day(-1) lowered liver triglyceride levels and improved liver markers such as alanine aminotransferase, NAFLD activity score, and fibrosis. Importantly, no changes in body temperature or food intake were observed. As potential treatment of NASH, mitochondrial uncouplers show promise for future development.

In this study, ABA triblock copolymers derived from 4,4′-(hexafluoroisopropylidene)-diphthalic anhydride-2,3,5,6-tetramethyl-1,4-phenylenediamine (6FDA-TeMPD; PI) and 3-hydroxy-1-adamantyl methacrylate (HAdMA) or 3,5-dihydroxy-1-adamantyl methacrylate (DHAdMA) have been synthesized via atom transfer radical polymerization to generate mechanically tough and thermally stable gas-separation membranes with composition-tunable transport properties. Due to the inherent thermodynamic incompatibility between the chemically dissimilar blocks, these two series of triblock copolymers appear microphase-separated. While the gas permeability coefficients of these triblock copolymer membranes are consistently lower than that of PI due to the reduced fractional free volume of HAdMA and DHAdMA, the solubility coefficients of the copolymers are higher than that of PI due presumably to specific interactions between the polar hydroxyl group(s) and penetrant gas molecules. These triblock copolymers synthesized from PI and hydrophilic adamantane derivatives constitute a new class of nanostructured polymeric materials possessing excellent thermomechanical properties in conjunction with designer gas-separation properties.
